摘 要:为进一步研究茯苓甘草汤有效成分和探讨其作用机制,分别利用中药系统药理学数据库与分析平台(TCMSP)和Genecards数据库筛选茯苓甘草汤的活性成分作用靶点和腹水的疾病靶点,将两者靶点取交集后利用Cytoscape软件构建“成分靶点”网络;采用String数据库构建PPI网络,拓扑分析获取关键靶点和成分;通过David数据库进行GO富集分析和KEGG通路富集分析;最后通过分子对接技术进一步佐证研究的可靠性。结果显示,茯苓甘草汤中活性成分共117种,可干预腹水相关靶点252个,GO富集519个,KEGG通路108条;其中,槲皮素、山奈酚、β-谷甾醇等可能为茯苓甘草汤治疗腹水的关键成分,PTGS2、ESR1、AR、BCL2、JUN等为潜在靶点;GO富集分析的生物功能涉及到凋亡负调节、细胞增殖、炎症反应等,KEGG通路富集分析涉及到TNF、PI3K-Akt、FoxO、HIF-1等信号通路;超过90%的分子对接结果都具有一定的结合活性。For further research on the effective ingredients and mechanism of Fuling Gancao decoction,the database and analysis platform of TCMSP and Genecards database were used to screen the active ingredient targets of Fuling Gancao decoction and the disease targets of ascites.After the intersection of the two targets,Cytoscape software was used to construct the“component-target”network;PPI network was constructed by String database,and key targets and components were obtained by topology analysis;GO enrichment analysis and KEGG pathway enrichment analysis were performed through David database;Finally,the reliability of this research was further confirmed by molecular docking technology.The results show that there are 117 active ingredients in Fuling Gancao decoction,which caninterfere with 252 ascites-related targets,519 GO enrichment and 108 KEGG pathways.Among them,quercetin,kaempferol,β-sitosterol might be the key components of Fuling Gancao decoction for the treatment of ascites,while PTGS2,ESR1,AR,BCL2,JUN are potential targets.The biological functions of GO enrichment analysis involve the negative regulation of death,cell proliferation,inflammation,etc.,and the KEGG pathway enrichment analysis involves TNF,PI3K-Akt,FoxO,HIF-1 and other signaling pathways.More than 90%of the molecular dockingresults have certain binding activity.
