摘 要:目的:探讨8周有氧运动训练对Nrf2敲除鼠运动能力和抗氧化系统的影响。方法:8周龄Nrf2敲除鼠(Nrf2 KO)和野生鼠(WT)各20只,随机分为安静组(n=10)和运动训练组(n=10),即分为野生安静组(WC组)、野生运动组(WE组)、敲除安静组(KC组)和敲除运动组(KE组)。运动组动物适应性训练1周后在标准小动物跑台上进行跑台运动。运动方案:坡度0°,12 m/min,60 min/天,5天/周,共持续8周。递增负荷强度测定小鼠运动能力。在最后一次运动48 h后,所有小鼠脱颈椎处死,取两侧股直肌下段,荧光比色法测定细胞ROS水平,RT-PCR测量Nrf2、CAT、NQO1和HO-1 mRNA表达量;Western blot法测定骨骼肌Nrf2、CAT、NQO1和HO-1蛋白表达。结果:1)WE组运动能力显著高于WC组,KE组显著高于KC组,KE、WE组间无显著性差异;2)KC、WC组间ROS无显著性差异,KE组显著低于WE组;3)KC组Nrf2、NQO1和CAT mRNA非常显著低于WC组,与WC组相比,WE组Nrf2、NQO1和HO-1 m RNA非常显著升高;4)除Nrf2外,KC组蛋白表达与WC组无显著性差异,KE组NQO1、HO-1和CAT较WE组均显著下降。结论:1)8周有氧运动通过Nrf2促进骨骼肌抗氧化酶的表达;2)8周有氧运动增强了小鼠运动能力,但并不依赖于Nrf2发挥作用。Objective: To investigate the effects of 8 weeks aerobic exercise training on the skeletal muscle antioxidant system and exercise capacity in Nrf2 knockout mice. Methods: 20 8-week-old Nrf2 knock out(KO) mice and 20 8-week-old wild mice(WT) were randomly assigned to a sedentary control group(n=10) and an exercise training group(n=10): Wild control(WC) group, wild exercise(WE) group, KO control(KC) group and KO exercise(KE) group.Mice in exercise groups underwent 0° slope treadmill running at speed of 12 m/min, 60 min/day,5 days per week for 8 weeks. The performance of exercise was measured by using increasing intensity. All the mice were sacrificed after 48 h of the last exercise. Fluorescence colorimetric assay was used to detect ROS levels in skeletal muscle of mice. The expression of Nrf2, NQO1,HO-1 and CAT m RNA in skeletal muscle of mice were determined by RT-PCR. The expression of Nrf2, NQO1, HO-1, CAT protein was detected by Western Blot. Results: 1) The exercise capacity of the WE group was significantly higher than that of the WC group, and the KE group was significantly higher than the KC group, while there was no significant difference between the KE group and the WE group;2) There was no significant difference in ROS between KC group and WC group, while KE group was significantly lower than WE group;3) The mRNA expression of Nrf2, NQO1 and CAT in the KC group were significantly lower than that in the WC group.Compared with the WC group, the mRNA expression of Nrf2, NQO1 and HO-1 in the WE group were significantly increased;4) In addition to Nrf2, the expression of protein in KC were no significant difference with the WC group, while the expression of NQO1, HO-1 and CAT in KE group were significantly lower than those of the WE group. Conclusions: 1) 8-week aerobic exercise promoted the expression of skeletal muscle antioxidant enzymes through Nrf2;2) The8-week aerobic exercise enhanced the exercise capacity, but did not rely on Nrf2.
