摘 要:为有效提高沙苑子总黄酮的得率,优选最佳提取工艺,并进行体外抗氧化活性研究。采用微波-超声波相结合的方法,在单因素实验结果基础上,筛选出影响总黄酮得率较显著的三个因素(乙醇浓度、微波时间和超声时间),以总黄酮得率为评价指标,采用响应面Box-behnken中心组合的方法进行考察。确定最佳提取工艺为乙醇浓度50%,微波时间130 s,超声时间47 min,得率可达到1.81%(17.78 mg/g);在此条件下研究其抗氧化活性,DPPH自由基清除率达到89%,IC_(50)为14.68μg/mL。该提取工艺稳定合理,提高了黄酮得率,且提取的黄酮具有较好的抗氧化活性。In order to effectively improve the extraction rate of total flavonoids from Astragalus complanatus, the optimal extraction technology was optimized and the antioxidant activity in vitro was studied. Using the method of combining microwave and ultrasonic wave, on the basis of single factor experimental results, three factors(ethanol concentration, microwave time and ultrasonic time) were selected to significantly affect the yield of total flavonoids. With mention of flavonoid yield as the evaluation index, the method of response surface Box-behnken center combination was used to investigate the results. With the optimum extraction process for ethanol concentration of 50%, microwave time 130 s, ultrasonic time 47 min, the yield can reach 1.81%(17.78 mg/g). Under these conditions, the scavenging rate of DPPH free radical reached 89% and IC_(50)was 14.68 μg/mL. The extraction process was stable and reasonable, the extraction rate was improved, and the extracted flavonoids had good antioxidant activity.
