摘  要:目的:优化光果甘草愈伤组织中甘草定提取条件,为后续细胞培养中成分检测提供方法。方法:愈伤组织干粉为材料,单因素试验基础上进行正交设计,考察液料比、乙醇浓度、温度和时间对甘草定提取率的影响。结果:液料比80∶1、乙醇浓度70%、60℃、100 min提取条件下光果甘草愈伤组织中甘草定提取率最高,为0.326%,RSD值为0.50%。结论:此工艺操作简便,稳定可行。Objective: To explore the extraction conditions glabridin from callus of Glycyrrhiza glabra L., and provide a method for glabridin detection during cell culture. Methods: Taking callus dry powder as material, based on the single factor experiment, the effects of liquid-solid ratio, ethanol concentration, temperature and time on the extraction rate of glabridin from callus dry powder were investigated by orthogonal experiment design.Results: The extraction rate of glabridin under the condition of 80∶1 liquid to solid ratio, 60% ethanol concentration, 50 ℃ extraction temperature, 60 min extraction time was the highest to 0.326%, and its RSD was 0.50%. Conclusion: The extraction processes were simple, stable and feasible.
