摘　　要：本试验旨在评价毕赤酵母表达系统生产的重组鸡CD40L对新城疫LaSota疫苗免疫效果的影响。首先构建表达鸡CD40L的重组毕赤酵母菌株,经纯化培养上清获得重组鸡CD40L。再分别用剂量为1.0μg/羽和0.1μg/羽的重组鸡CD40L和新城疫LaSota疫苗联合免疫7日龄SPF鸡。于免疫后14、21、28 d和35 d通过血凝血抑试验检测血清中新城疫抗体水平,用商品试剂盒检测血清超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活力以及丙二醛(MDA)含量。结果显示,重组鸡CD40L联合新城疫LaSota疫苗免疫不能增强疫苗诱导产生的血清抗体水平,不能提高鸡血清SOD活性,但能提高血清GSH-Px活性以及降低血清MDA含量。因此,在本试验所设剂量(1.0μg/羽和0.1μg/羽)下,重组鸡CD40L对疫苗无明显的免疫增效作用,但能够增强鸡的抗氧化能力。This study aimed to evaluate the effect of recombinant chicken CD40 L produced in Pichia pastoris expression system on Newcastle disease LaSota vaccine. First, recombinant Pichia pastoris strain was constructed to express chicken CD40 L, and the resulting recombinant chicken CD40 L was purified from culture supernatant. Then, 7-day-old SPF chickens were immunized with recombinant chicken CD40 L and Newcastle disease LaSota vaccine at doses of 1.0 μg per chicken and 0.1 μg per chicken, respectively. The levels of Newcastle disease antibodies in sera were determined at 14, 21, 28 days and 35 days post immunization by HA/HI test, and the superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) vitality and malondialdehyde(MDA) content in sera were detected by commercial kits. The results showed that the recombinant chicken CD40 L in combination with Newcastle disease LaSota vaccine did not increase the level of serum antibodies to Newcastle disease vaccine, nor the level of serum SOD activity, but increased the serum activity of GSH-Px and reduced the serum content of MDA. Thus, at the dose 1.0 μg per chicken and 0.1 μg per chicken set in this study, recombinant chicken CD40 L has no obvious synergistic effect on Newcastle disease LaSota vaccine, but can enhance the antioxidant capacity in chicken.
