摘 要:目的研究红毛五加叶水提取物对大鼠脑缺血损伤的保护作用及机制。方法将雄性SD大鼠随机分为假手术组,脑缺血模型组,银杏叶片组,红毛五加叶水提取物高、中、低剂量组。银杏叶片组灌胃银杏叶片40 mg/kg,红毛五加叶水提取物高、中、低剂量组分别灌胃红毛五加叶水提取物20、10、5 g/kg,假手术组和脑缺血模型组灌胃等体积的生理盐水(2 mL/kg),1次/d,连续7 d。除假手术组外,其余各组采用夹闭或结扎双侧颈总动脉的方法建立大鼠脑缺血损伤模型。检测大鼠学习记忆能力、脑指数、脑含水量、脑毛细血管通透性及脑组织腺苷三磷酸、乳酸、超氧化物歧化酶、丙二醛水平。结果与假手术组比较,脑缺血模型组大鼠学习记忆能力显著下降(P<0.05)。脑指数、脑含水量、脑毛细血管通透性及脑组织乳酸、丙二醛水平显著增加(P<0.01),腺苷三磷酸、超氧化物歧化酶水平显著降低(P<0.01)。与脑缺血模型组比较,红毛五加叶水提取物高、中、低剂量组上述指标异常改变均有明显改善。结论红毛五加叶水提取物对大鼠脑缺血损伤具有保护作用,其机制与提高抗氧化能力有关。Objective To study the protective effects and mechanism of water extracts of Acanthopanax giraldii Harms leafs on cerebral ischemia injury in rats.Metholds Male adult SD rats were randomly divided into the sham opertion group,the cerebral ischemia model group,Ginkgo bioba tablet group,and water extracts of Acanthopanax giraldii Harms leafs high,medium,low dose group.Rats in the Ginkgo bioba tablet group were intragastrically given Ginkgo bioba tablet 40 mg/kg,water extracts of Acanthopanax giraldii Harms leafs high,medium and low dose groups were intragastrically given water extracts of Acanthopanax giraldii Harms leafs 20,10 and 5 g/kg respectively;while rats in the sham operation group and cerebral ischemia model group were intragastrically given equal volume of nomal saline(2 mL/kg),once a day,for 7 d.All rats were induced for cerebral ischemia injury model by bilateral common carotid artery occlusion method except for the sham operation group.Learning-memory ability was observed,the cerebral index,cerebral water content,cerebral capillary permeability and the levels of adenosine triphosphate,lactic acid,superoxixde dismutase,malondiadehyde in brain tissue were measured in rats.Results Compared with the sham operation group,the learning and memory ability of rats in the cerebral ischemia model group decreased significantly(P<0.05).Cerebral index,cerebral water content,cerebral capillary and the levels of lactic acid and malondialdehyde in the brain tissue increased significantly(P<0.01),while the levels of adenosine triphosphate and superoxide dismutase decreased significantly(P<0.01).Compared with the cerebral ischemia model group,the abnormal changes of the above indexes in the high,medium and low dose groups of water extracts of Acanthopanax giraldii Harms leafs were significantly improved.Conclusion Water extracts of Acanthopanax giraldii Harms leafs has protective effects on cerebral ischemia injury in rats,and its mechanism may be related to an increased antioxidant capacity.
