摘 要:本文旨在探究新疆荒漠肉苁蓉毛蕊花苷部位对慢性全脑低灌注(CCH)致小鼠认知功能障碍的影响。于双侧颈总动脉外套以内径0.19 mm的不锈钢微弹簧复制小鼠CCH模型,甲苯胺蓝O灌注染色考察脑血流灌注水平,Morris水迷宫考察小鼠空间学习记忆功能,于饮水中补充毛蕊花苷提取物连续干预4个月,比色法测定海马组织乙酰胆碱酯酶(AChE)和超氧化物歧化酶(SOD)活性以及丙二醛(MDA)水平。结果表明,造模后4个月,小鼠表现出明显的认知功能障碍,于饮水中补充毛蕊花苷提取物(0.16~0.64 g/L)可明显改善CCH小鼠认知功能障碍,逆转模型小鼠脑组织AChE活性和MDA水平的升高,同时上调模型小鼠脑组织SOD活性。本研究证实于饮水中补充毛蕊花苷提取物,可明显改善CCH小鼠认知功能障碍,或与其改善低灌注致脑组织氧化损伤相关。The purpose of this study is to explore the protective effect of verbascoside components from desert Cistanche in Xinjiang against the cognitive dysfunction induced by chronic cerebral hypoperfusion(CCH)in mice.The mice are subjected to bilateral common carotid artery stenosis using external microcoils with inner diameters of 0.19 mm to copy CCH model.The cerebral blood perfusion level is investigated by Toluidine blue O perfusion staining.The spatial learning and memory function is evaluated by Morris water maze.Supplementing verbascoside in drinking water for continuous intervention for 4 months,the activities of acetylcholinesterase(AChE)and superoxide dismutase(SOD)and the malondialdehyde(MDA)level in hippocampal tissues are detected by colorimetric.Four months after modeling,the mice show obvious cognitive dysfunction.Supplementing verbascoside(0.16-0.64 g/L)in drinking water improves the cognitive dysfunction of CCH mice.Moreover,verbascoside also reverses the increase of AChE activity and MDA level and the decrease of SOD activity in hippocampus tissue in CCH mice.These results demonstrate that the verbascoside components can ameliorate the cognitive impairment induced by CCH,which may be partially due to its anti-oxidative properties.
