摘　　要：目的建立HPLC同时测定咳清胶囊中吗啡、磷酸可待因和岩白菜素含量的方法。方法采用Ultimate■AQ-C_(18)色谱柱(4.6 mm×250 mm,5μm),乙腈(A)-0.1%磷酸溶液(B)为流动相,梯度洗脱,流速为1.0 mL·min^(-1),柱温为35℃,检测波长为210 nm,进样量10μL。结果吗啡、磷酸可待因和岩白菜素分别在20.0182~320.2912μg·mL^(-1)(r=0.9999)、4.0310~64.4966μg·mL^(-1)(r=1.0000)、19.7600~316.1600μg·mL^(-1)(r=1.0000)内线性关系良好;平均回收率(n=6)分别为98.33%(RSD=2.72%),98.72%(RSD=2.77%)和98.54%(RSD=1.06%)。结论该检测方法简便、准确、重复性好,可用于咳清胶囊中吗啡、磷酸可待因和岩白菜素3种有效成分的同时测定,也可为其质量控制提供参考。OBJECTIVE To establish a HPLC method for the simultaneous determination of morphine,codeine phosphate and bergenin in Keqing capsules.METHODS Ultimate■AQ-C_(18)(4.6 mm×250 mm,5μm)column was used;mobile phase was acetonitrile(A)-0.1%phosphoric acid(B)with gradient elution;flow rate was 1.0 mL·min^(-1);column temperature was 35℃;detection wavelength was 210 nm and the injection volume was 10μL.RESULTS Good linear relationship was observed for morphine,codeine phosphate and bergenin in the range of 20.0182-320.2912μg·mL^(-1)(r=0.9999),4.0310-64.4966μg·mL^(-1)(r=1.0000),and 19.7600-316.1600μg·mL^(-1)(r=1.0000),respectively.The average recoveries(n=6)were 98.33%(RSD=2.72%),98.72%(RSD=2.77%)and 98.54%(RSD=1.06%),respectively.CONCLUSION The established HPLC method is simple,accurate,repeatable and can be used for the determination of morphine,codeine phosphate and bergenin in Keqing capsule.It can also provide reference for its quality control.
