摘 要:为了丰富生防微生物菌种资源库,从杨山牡丹的根、茎、叶中共分离出211株内生细菌,经初筛和复筛,有6株菌株(YMG34、YMJ74、YMJ78、YMJ141、YMJ143、YMY13)对牡丹黑斑病菌(Alternaria suffruticosae)、黄斑病菌(Phyllosticta commonsii)、腔孢叶斑病菌(Hainesia lythri)、灰霉病菌(Botrytis paeoniae)和北方根结线虫(Meloidogyne hapla)表现出稳定的拮抗活性。拮抗实验结果表明,6株内生菌处理能使牡丹病原真菌菌丝生长扭曲,出现泡囊结构以及局部膨大。经生理生化实验并结合16S rDNA及gyrB基因序列分析,YMG34菌株被鉴定为巨大芽孢杆菌(Bacillus megaterium);其余5个菌株被鉴定为耐盐芽孢杆菌(B.halodurans),结果表明6株内生细菌菌株均具有防治牡丹病害的潜力。In order to enrich the resource library of biocontrol microbial species,211 endophytic bacterial strains were isolated from the root,stem and leaf of Paeonia ostii.After initial screening and rescreening,six strains(YMG34,YMJ74,YMJ78,YMJ141,YMJ143 and YMY13)showed the stable antagonistic activity to Alternaria suffruticosae,Phyllosticta commonsii,Hainesia lythri,Botrytis paeoniae and Meloidogyne hapla.The antagonism test showed that the mycelia of pathogenic fungi treated with six endophytic bacteria showed abnormal growth,twist,bubble deformation structures and differential expansion.Based on physiological and biochemical characters,16S rRNA and gyrB gene sequence analysis,YMG34 was identified as Bacillus megaterium,and the other strains were identified as B.halodurans.The results demonstrated the biocontrol potential of 6 endophytic bacteria against peony diseases.
