摘 要:目的:通过观察知母多糖对热证模型大鼠肝脏代谢的干预效果,探讨知母多糖的药性归属,为阐明中药知母寒性药性的内涵提供现代科学依据。方法:SD大鼠随机分为对照组、模型组和给药组。给予模型组和给药组大鼠优甲乐进行甲亢热证造模,造模期间给予给药组大鼠知母多糖水溶液,对照组和模型组给予等量蒸馏水,连续15 d。给药结束后,通过UPLC-TOF/MS对3组大鼠肝脏组织进行代谢物定量分析,通过与HMDB数据库比对定性代谢物,运用生物信息学方法分析并比较模型组与给药组肝脏代谢通路的变化情况。结果:PCA结果显示3组代谢物主成分差异显著。经OPLS-DA分析,模型组与空白组比对共有11个显著差异代谢物,给药组与模型组比对共有10个显著差异代谢物。通路分析结果显示,模型组甘油磷脂、亚油酸代谢通路明显改变,而给药组中磷脂酰胆碱(PC)、磷脂酰乙醇胺(PE)和磷脂酸(PA)相对模型组回调,逆转了模型组甘油磷脂与亚油酸代谢通路的变化。结论:知母多糖可逆转典型热证模型的代谢通路变化,表明知母多糖药性为寒性,可通过肝脏物质能量代谢的改变发挥药效。Objective:To investigate the medical property of Rhizoma Anemarrhenae polysaccharide(RAPS)by observing its intervention effect on liver metabolism in rats with hyperthyroidism of heat syndrome,thus to provide modern scientific evidence for the connotation of the cold nature of Rhizoma Anemarrhenae.Methods:24 male SD rats were randomly divided into the control group,the model group and the RAPS group. The hyperthyroidism model of heat syndrome was prepared by Euthyrox. During the modeling,different interventions were accordingly given to each group for 15 days. After the treatment,the quantitative metabolic analysis of livers was conducted by UPLC-TOF/MS in the three groups,the metabolites were determined by alignment in the HMDB database,and the variation of liver metabolic pathway between the paired-comparison groups was analyzed in a way of bioinformatics. Results:The PCA results indicated that the principle components of the three groups were visibly discrepant. By analysis of OPLS-DA,the model group possessed 11 significantly differentiated metabolites compared with the control group,and the RAPS group possessed 10 significantly differentiated metabolites compared with the model group. The pathway analysis showed that glycerophospholipid metabolism and linoleic acid metabolism were significantly changed in the model group;the tendency of glycerophospholipid metabolism and linoleic acid metabolism were reversed in the RAPS group by call-back of phosphatidylcholine(PC),phosphatidylethanolamine(PE)and phosphatidylic acid(PA)compared with the model group. Conclusion:RAPS can reverse the significant metabolic pathway change in the liver of the classic model of heat syndrome,which demonstrates that RAPS possesses a cold nature and it can take effect by altering the energy metabolism in liver.
