耳针治疗对慢性心力衰竭大鼠血清BNP、ET-1和IL-6的影响

摘　　要：目的:探讨耳针治疗对慢性心力衰竭(CHF)模型大鼠血清BNP、ET-1和IL-6的影响,为临床采用耳针治疗慢性心力衰竭提供依据。方法:选取40只健康雄性SD大鼠,依据随机数字表随机分为对照组、模型组、假耳针治疗组和耳针治疗组。除对照组外,其余3组采用腹腔注射阿霉素的方法制作CHF模型。造模成功后,对照组和模型组不予任何治疗;耳针治疗组和假耳针治疗组分别予以耳针和假耳针治疗2个月。对比干预前后各组大鼠血清脑钠肽(brain natriuretic peptide,BNP)、内皮素-1(endothelin-1,ET-1)和白细胞介素-6(interleukin-6,IL-6)水平的变化情况,并观察超声心动图指标。结果:治疗后,各组实验大鼠超声心动图指标比较,模型组、假耳针治疗组和耳针治疗组较对照组LVIDd、LVIDs水平升高(P <0.05),LVFS和LVEF水平降低(P <0.05)。模型组、假耳针治疗组和耳针治疗组血清BNP、ET-1和IL-6含量较对照组升高(P <0.05);与模型组比较,假耳针治疗组和耳针治疗组血清BNP、ET-1和IL-6含量降低(P <0.05);与假耳针治疗组比较,耳针治疗组血清BNP、ET-1和IL-6含量降低更明显(P <0.05)。结论:耳针治疗对慢性心衰模型大鼠有治疗作用,可降低血清BNP、ET-1和IL-6水平,改善左心室舒缩功能。Objective:To investigate the effects of auricular acupuncture on serum BNP,ET-1 and IL-6 in the rat model of chronic heart failure(CHF),thus to provide evidence for the treatment of CHF with auricular acupuncture. Methods:40 healthy male SD rats were randomly divided into the control group,the model group,the sham auricular acupuncture group and the auricular acupuncture group. CHF model was established by intraperitoneal injection of Adriamycin. The intervention duration was 2 months. The serum levels of brain natriuretic peptide(BNP),endothelin-1(ET-1)and interleukin-6(IL-6)were compared between before and after the intervention in the two groups. Echocardiographic indicators were also observed in the two groups. Results:Compared with those in the control group,LVEDd and LVIDs were increased(P < 0. 05),whereas LVFS and LVEF were decreased in the model group,the sham auricular group and the auricular group after the treatment(P < 0. 05). The serum contents of BNP,ET-1 and IL-6 were increased in the model group,the sham auricular acupuncture group and the auricular acupuncture group compared with those in the control group after the treatment(P < 0. 05);compared with those in the model group,the serum contents of BNP,ET-1 and IL-6 were decreased in the sham auricular acupuncture group and the auricular acupuncture group(P < 0. 05),and the decreases in the auricular acupuncture group were more significant than those in the sham auricular acupuncture group(P < 0. 05). Conclusion:Auricular acupuncture therapy has a significant effect on CHF model rats,it can reduce serum levels of BNP,ET-1 and IL-6 and improve the functions of contraction and diastole of left ventricle.
