猪HIF-1α基因启动子区及编码区的生物信息学分析

摘  要:利用生物信息学方法,对NCBI中公布的猪缺氧诱导因子HIF-1α基因的5′端上游2000 bp长度的候选启动子区进行了核心启动子及转录结合位点的预测;同时对其所编码蛋白的理化性质、亚细胞定位、跨膜结构域、信号肽、高级结构及进化关系进行了分析。结果显示:猪HIF-1α基因5′端上游存在潜在启动子区,存在CpG岛,存在多个转录因子结合位点,编码的蛋白为亲水性蛋白;蛋白质等电点为5.11,共由824个氨基酸组成,含量最高的为亮氨酸。HIF-1α蛋白主要定位于细胞核内,不存在跨膜结构域,为非跨膜蛋白,不存在信号肽,二级结构主要为α螺旋及无规卷曲。此外通过对猪HIF-1α基因序列的同源性比较发现,与猪HIF-1α基因同源性最近的是绵羊,同源性最远的是原鸡。此研究为猪HIF-1α基因结构和功能的探索提供了一定的理论依据。Using the method of bioinformatics,the core promoter and transcription binding site are predicted in the candidate promoter region with a length of 2000 bp upstream of the 5′end of the pig hypoxia inducible factor HIF-1αpublished in NCBI.The physicochemical properties,subcellular localization,transmembrane domain,signal peptide,advanced structure and evolutionary relationship of the protein encoded by HIF-1αare analyzed.The results show that there is a potential promoter region at the upstream of 5′end of pig's HIF-1α.There are CpG islands and multiple transcription factor binding sites.The protein encoded by HIF-1αis a hydrophilic protein with an isoelectric point of 5.11.It is composed of 824 amino acids,and the highest content is leucine.HIF-1αprotein is mainly located in the nucleus,and there is no transmembrane domain,so it is a non transmembrane protein.There is no signal peptide in the HIF-1αprotein,and its secondary structure is mainlyα-spiral and random curl.In addition,through the comparison of HIF-1αsequence homology,it is found that sheep has the closest homology with pig's HIF-1αgene and the original chicken has the furthest homology.In conclusion,the study provides a theoretical basis for the exploration of structure and function of pig's HIF-1αgene.
