摘 要:目的:冬季奥运会比赛在冷环境下进行。冷环境对运动耐力的影响及其调节机制的相关研究较少。钙泵调节蛋白(sarcolipin,SLN)在骨骼肌产热维持体温平衡中具有重要作用。拟通过小鼠在冷环境下运动,探究冷暴露对小鼠骨骼肌中SLN表达和运动耐力的影响。方法:30只8周龄健康雄性C57BL/6N小鼠随机分为3组,每组10只:安静对照组(Con)、常温运动组(Exe)和低温运动组(Cold+Exe)。Con组与Exe组置于(25±1)℃的室温环境下,Cold+Exe组在运动能力测试时置于(4±1)℃冷环境中,Exe组与Cold+Exe组进行逐增负荷至力竭的跑台运动,跑动时间和距离作为运动耐力的评定指标。运动后即刻取小鼠血液、腓肠肌和股四头肌,对血清肌酸激酶(CK)、肌浆网中Ca;、骨骼肌ATP含量、SLN及Ca;调节蛋白表达以及糖脂能量代谢相关指标等进行测定。结果:与Exe组相比,Cold+Exe组:(1)小鼠跑动时间和跑动距离均显著降低;(2)肌浆网Ca;浓度显著下降;(3)骨骼肌糖脂代谢的Pparδ、Acadm、Glut4的mRNA表达和线粒体复合物Ⅳ活性增加;(4)肌浆网上Ca;释放通道RyR表达下降,钙泵SERCAⅡ和SLN表达显著增加。结论:冷环境(4℃)下,运动可降低机体的运动耐力;骨骼肌SLN表达增加,导致钙泵SERCA解偶联和消耗ATP无效工作循环产生热量,从而使运动系统供能不足,可能是造成运动耐力降低的重要原因。Objective:The Olympic Winter Games are held in a cold environment. There is few research on the effects of cold exposure on exercise endurance and its regulatory mechanism. SERCA regulatory protein(sarcolipin,SLN)plays an important role in skeletal muscle thermogenesis to maintain body temperature homeostasis. This research aims to explore the effects of cold exposure on the expression of Skeletal Muscle SERCA Regulatory Protein SLN and exercise endurance in mouse skeletal muscle. Methods:Thirty 8-week-old healthy male C57BL/6N mice were randomly divided into 3 groups with 10 mice in each group:control group(Con),normal temperature exercise group(Exe)and low temperature exercise group(Cold+Exe).The Con group and the Exe group were placed in a room temperature environment of(25±1)℃,the Cold+Exe group was placed in a(4±1)℃ cold environment during the exercise ability test,and the Exe group and the Cold+Exe group were taking Treadmill exercise with gradually increased load until exhaustion. Running time and distance were used as indicators of exercise endurance. The blood,gastrocnemius muscle and quadriceps femoris muscle of mice were collected immediately after exercise,and serum creatine kinase(CK),Ca;in sarcoplasmic reticulum,ATP content of skeletal muscle,expression of SLN and Ca;regulatory proteins,and related indicators of glycolipid energy metabolism were measured. Results:Compared with the Exe group,the Cold+Exe group:(1)the running time and running distance of the mice were significantly shorter;(2)the concentration of Ca;in the sarcoplasmic reticulum significantly decreased;(3)the mRNA expression of Pparδ,Acadm and Glut4 and the activity of mitochondrial complex IV increased;(4)the expression of Ca;release channel RyR on the sarcoplasmic reticulum decreased,and the expressions of calcium pump SERCAⅡ and calcium pump regulatory protein SLN significantly increased.Conclusion:Exercise in a cold environment(4 ℃)can reduce exercise endurance.
