摘 要:目的探讨芍药苷对由异丙肾上腺素(ISO)引起的大鼠心肌重构的影响及机制。方法32只SD大鼠随机分为4组,分别为对照组、模型组、高剂量组[芍药苷80 mg/(kg·d)]和低剂量组[芍药苷40 mg/(kg·d)]。模型组和高、低剂量组连续皮下注射ISO(5 mg/kg)1周,对照组注射等容量生理盐水;造模同日起,高、低剂量组灌胃芍药苷,模型组、对照组灌胃等量生理盐水,连续灌胃14 d;末次给药后,记录大鼠心电图、测定心率;测定全心指数及左心室指数;测定心肌组织SOD、MDA、GSH-Px及Ⅰ型胶原含量;并进行心肌组织病理学观察。结果高剂量芍药苷可明显减慢大鼠心率,降低全心指数、左心室指数和心肌组织MDA、Ⅰ型胶原蛋白含量,提高心肌SOD、GSH-Px活性,减轻心肌纤维化,对大鼠心肌重构具有一定的干预作用。结论芍药苷对ISO致大鼠心肌重构具有一定的干预作用,其作用机制可能与提高抗氧化能力有关。Objective:To study the effect and mechanism of paeoniflorin on myocardialremodeling induced by isoproterenol(ISO)in rats.Methods:Thirty-two SD rats were randomly divided into four groups:control group,model group,high dose group[80 mg/(kg·d)paeoniflorin]and low dose group[40 mg/(kg·d)paeoniflorin].The model group,high-dose group and low-dose group were subcutaneously injected with ISO(5 mg/kg)for one week,while the control group was injected with the same volume of normal saline;From the same day of modeling,the high-dose and low-dose groups were gavaged with paeoniflorin,and the model group and control group were gavaged with the same amount of normal saline for14 days;After the last administration,ECG and heart rate were recorded;The whole heart index and left ventricular index were measured;The contents of SOD,MDA,GSH-Px and type I collagen in myocardium were measured,and the myocardial histopathology was observed.Results:Paeoniflorin could significantly slow down the heart rate,decrease the whole heart index,left ventricular index,myocardial MDA and type I collagen content.Paeoniflorin could improve the activity of myocardial SOD and GSH-Px,and reduce myocardial fibrosis.Paeoniflorin had a certain intervention effect on myocardial remodeling in rats.Conclusion:Paeoniflorin has a certain intervention effect on myocardial remodeling induced by ISO in rats,and its mechanism may be related to the improvement of antioxidant capacity.
