摘　　要：目的:研究建立不同激发方式的流式细胞分选仪一体化管理模式,探讨其应用效果。方法:针对流式分选仪的组成、工作原理、环境要求、人员要求、培训交流、操作管理及注意事项,探讨激光在空气中激发和在石英杯中激发两种流式细胞分选仪的使用及管理方法。结果:总结出一套日常维护、运行及培训的开放一体化管理模式,仪器每日稳定运行较一体化管理之前的8 h提高到10~12 h,运行机时数>1500 h/年,分选后细胞活性>90%,分选细胞纯度>99%,污染率<1%,样品堵塞<2%。结论:不同激发方式的流式细胞分选仪一体化管理模式能够加强仪器运行及维护管理,可使仪器使用者快速熟悉操作分选仪,延长分选仪使用寿命,提高分选仪使用率,为获得高纯度、高活性和高得率细胞提供保证,为科学研究、实验教学及技术开发提供更优质服务。Objective:To study and establish the integrated management mode of flow sorter with different excitation modes,and explore its application effect.Methods:In view of the composition,working principle,environment requirements,personnel requirements,training and exchange,operation management and matters needing attention of the flow sorter,the usage and management of two kinds of flow sorter with laser excitation in air and laser excitation in quartz cell were explored.Results:A set of open and integrated management mode of daily maintenance,operation and training was summarized.The daily stable operation of the sorter was increased to 10-12 hours compared with 8 hours before the integrated management,the operating hours were more than 1,500hours/year,the post-sorting cell activity was>90%,the sorted cell purity was>99%,the contamination rate was<1%,and the sample clogging was<2%.Conclusion:The integrated management mode of flow sorters with different excitation modes can strengthen the operation and maintenance management of the sorter,enable the users quickly familiar with operation of the sorter,prolong the service life of sorter,improve the utilization rate of the sorter,guarantee for obtaining cells of high purity,high activity and high yield,and provide better service for scientific research,experiment teaching and technology development.
