摘 要:目的观察青紫颗粒对Arthus反应大鼠、兔皮肤损伤的保护作用并探讨其作用机制。方法通过肌内注射卵蛋白-弗氏完全或不完全佐剂,皮内注射1%卵蛋白溶液,复制Arthus反应大鼠和家兔模型。采用卵蛋白-弗氏完全佐剂致敏,1%卵蛋白溶液攻击,复制Arthus反应兔模型。观察抗原攻击后皮肤反应程度、局部红肿斑点最大直径,检测血清IgA、IgG、CIC等指标的含量,并观察皮肤病理改变。结果在抗原攻击后4 h,青紫颗粒低剂量可减小大鼠背部斑点直径;抗原攻击后24 h,中、高剂量可减小背部斑点直径,中剂量可减轻皮肤反应程度。青紫颗粒各剂量可降低大鼠血清IgA、IgG、NO含量;中、高剂量可降低CIC含量;高剂量可降低血清IL-6、TNF-α含量。青紫颗粒各剂量可减轻大鼠皮肤组织静脉血管淤血扩张、出血情况。在抗原攻击后2、3、4、5 h,青紫颗粒高剂量可减小兔背部皮肤斑点直径;高、低剂量可减轻皮肤反应程度。青紫颗粒中剂量可降低兔血清IgG含量,中、低剂量组可降低血清CIC含量。青紫颗粒各剂量组均可减轻兔皮肤血管扩张,低剂量组可缓解皮肤水肿。结论青紫颗粒对Arthus反应动物模型皮肤损害具有一定保护作用,该作用可能与其降低血清IgA、IgG、CIC、IL-6、TNF-α、NO水平有关。Objective To determine the protect effect of Qingzi granules on skin damage in rats and rabbits with Arthus response and related mechanism of action.Methods The Arthus response rat and rabbit model was established by intramuscular injection of ovalbumin-Freund complete or incomplete adjuvant and intradermal injection of 1%ovalbumin solution.The degree of skin reaction at different time points after the antigen attack was observed,and the maximum diameter of swelling and the serum levels of IgA,IgG,CIC,and so on were detected,and the skin pathological changes were examined.Results Qingzi granules low-dose decreased the diameter of the back swollen spots at 4 h after antigen attack in rats.Qingzi granules at middle and high-dose significantly decreased the diameter of the back swollen spots at 24 h after the antigen attack.Qingzi granules at middle dose reduced the skin reaction.All doses of Qingzi granules lowered the serum levels of IgA,IgG and NO in rats;Qingzi granules middle and high-dose decreased the serum level of CIC;while high-dose decreased the serum levels of IL-6 and TNF-α.All doses of Qingzi granules alleviated the nuclear fragmentation,venous congestion,vasodilation and expansion,and hemorrhage in rats.At 2,3,4,and 5 hours after the antigen attack,Qingzi granules high-dose decreased the diameter of swollen spots on the back in rabbits;while high and low-dose reduced the skin reaction.Qingzi granules middle-dose lowered the serum IgG level in rabbits,and middle and low-dose decreased the serum CIC content.All doses of Qingzi granules alleviated the skin vasodilation,and low-dose Qingzi granules relieved the skin edema in rabbits.Conclusion Qingzi granules can alleviate the skin damage in rats and rabbits with Arthus response,and its mechanism of action may be related to reducing the levels of IgA,IgG,CIC,IL-6,TNF-αand NO.
