摘 要:目的探究黄连-干姜对糖尿病大鼠脑脊液中的Aβ42、p-tau、Ach含量动态变化的影响。方法腹腔注射1%STZ 55 mg·kg^(-1)复制大鼠糖尿病模型,将糖尿病大鼠按照血糖和体质量随机分成模型组、黄连-干姜组、二甲双胍组,连续给药75 d,并动态监测各组糖尿病大鼠空腹血糖;每隔15 d应用小脑延髓池穿刺法采集脑脊液,用酶联免疫吸附法动态测定各组药物对大鼠脑脊液中的Aβ42、p-tau、Ach含量变化,以及建模75 d大鼠后脑组织内Aβ42的含量。结果与空白组比较,模型组大鼠空腹血糖显著升高,模型稳定;大鼠脑脊液中Aβ42的含量不断下降,在用药30 d时含量显著降低;脑脊液中Ach的含量在用药30、60、75 d均较低;脑脊液中p-tau的含量在用药60、75 d时显著升高;脑组织中Aβ42的含量显著增加。而黄连-干姜对此变化趋势具有调节作用,与模型组比较,黄连-干姜组大鼠用药60、75 d空腹血糖显著降低;脑脊液中Aβ42的含量在用药30、60、75 d显著升高;脑脊液中Ach的含量在用药30 d显著升高;脑组织中Aβ42的含量显著减少。结论黄连-干姜可改善糖尿病大鼠空腹血糖,降低脑组织中Aβ42的含量,提高脑脊液中Aβ42、Ach的含量。Objective To determine the effect of Coptis-dried ginger on the dynamic changes of Aβ42,p-tau and Ach content in the cerebrospinal fluid of diabetic rats.Methods The rats were intraperitoneally injected 1%STZ 55 mg·kg^(-1) to create a diabetic model.The diabetic rats were randomly divided into a model group,a Coptis-dried ginger group and a metformin group according to their blood glucose and body weight.The rats were administered continuously for 75 d,and the fasting blood glucose of each group was dynamically monitored.At the same time,the cerebrospinal fluid was collected every 15 d,and the changes of Aβ42,p-tau and Ach content in the cerebrospinal fluid of rats treated by drugs in each group,as well as the content of Aβ42 in the brain tissue 75 d after the model establishment were determined dynamically by ELISA.Results Compared with the control group,fasting blood glucose was significantly increased in model group and the model was stable.The content of Aβ42 in cerebrospinal fluid of rats decreased continuously,and decreased significantly after 30 d of medication.The content of Ach in cerebrospinal fluid was low at 30,60 and 75 d.The content of p-tau in cerebrospinal fluid increased significantly at 60 and 75 d after the treatment.The content of Aβ42 in brain tissue was significantly increased.Compared with the model group,fasting blood glucose of rats in Coptis-dried ginger group was significantly decreased at 60 and 75 d after the treatment.The content of Aβ42 in cerebrospinal fluid increased significantly at 30,60 and 75 d after the treatment.Ach content in cerebrospinal fluid increased significantly 30 d after the treatment.The content of Aβ42 in brain tissue was significantly decreased.Conclusion Coptis-dried ginger can decrease the fasting blood glucose and Aβ42 content in the brain tissue and increase the content of Aβ42 and Ach in the cerebrospinal in diabetic rats.
