摘 要:目的建立山豆根药材UPLC-UV指纹图谱和同时测定其中三叶豆紫檀苷、高丽槐素、山豆根素含量的方法。方法采用Waters Acquity UPLC HSS T3色谱柱(2.1 mm×100 mm,1.8μm),以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,检测波长为203 nm,流速为0.3 mL·min^(-1),柱温为25℃,进样量为1μL,建立山豆根药材UPLC-UV指纹图谱分析方法及同时测定三叶豆紫檀苷、高丽槐素、山豆根素含量的方法,测定16批山豆根药材的指纹图谱及3种成分的含量。结果指纹图谱及含量测定方法学验证均良好,16批山豆根药材UPLC指纹图谱的相似度在0.926~0.989,确定10个共有峰,三叶豆紫檀苷含量为0.056%~0.226%,高丽槐素含量为0.04%~0.151%,山豆根素含量为0.163%~0.585%。结论本研究为山豆根整体质量评价提供了新的方法,可用于山豆根药材质量控制。Objective To establish the UPLC-UV fingerprint of Sophora Tonkinensis and simultaneously determine the content of trifolirhizin,sophoranone,and maackiain.Methods Waters Acquity UPLC HSS T3 column(2.1 mm×100 mm,1.8μm)was used with the mobile phase consisting of acetonitrile-0.1%phosphoric acid at a flow rate of 0.3 mL·min^(-1) for gradient elution.The column temperature was 25℃.The detection wavelength was 203 nm,and the injection volume was 1μL.The fingerprints of 16 batches of Sophora Tonkinensis and the content of trifolirhizin,sophoranone and maackiain were determined.Results The fingerprints and content determination methods were well verified.The similarity of UPLC fingerprints of the 16 batches of Sophora Tonkinensis was between 0.926 and 0.989,and 10 common peaks were calibrated.The content of trifolirhizin,maackiain,and sophoranone was 0.056%~0.226%,0.04%~0.151%,and 0.163%~0.585%.Conclusion This study provides a new method for the overall quality evaluation of Sophora Tonkinensis,which can be used for the quality control of Sophora Tonkinensis.
