摘  要:目的建立超高效液相色谱法测定胆木不同部位(根、枝、皮、叶)中异长春花苷内酰胺含量。方法样品采用75%甲醇超声提取;选用Waters BEH C_(18)色谱柱(100 mm×2.1 mm,1.7μm);流动相为乙腈(A)-纯水(B),梯度洗脱;流速0.4 mL·min^(-1);柱温40℃;进样量3μL;检测波长227 nm。结果异长春花苷内酰胺在0.11~85.60μg·mL^(-1)与峰面积线性关系良好(R^(2)=0.9991),平均加样回收率为99.77%,RSD为0.13%。异长春花苷内酰胺在根中含量最高(11.42~13.38 mg·g^(-1)),其次为叶(2.03~5.00 mg·g^(-1)),在枝中含量相对较低(1.57~3.79 mg·g^(-1))。结论该方法快捷、灵敏、准确、可靠,可用于胆木中异长春花苷内酰胺含量的测定,为胆木的质量控制提供参考。Objective To determine the content of isovincoside lactone in the root,branch,bark and leaf of Nauclea officinalis Pierrc ex Pitard by UPLC-PDA.Methods The samples were extracted by ultrasound with 75%methanol.The analysis was performed on a Waters BEH C_(18) chromatographic column(100 mm×2.1 mm,1.7μm)with gradient elution of acetonitrile-water at the flow rate of 0.4 mL·min^(-1).The column temperature was 40℃,the injection volume was 3μL,and the detection wavelength was 227 nm.Results There was a good linear relationship between the peak areas and the concentration of isovincoside lactone at 0.11~85.60μg·mL^(-1)(R^(2)=0.9991).The average recovery was 99.77%with the RSD of 0.13%.The content of isovincoside lactone in the root was the highest(11.42~13.38 mg·g^(-1)),followed by that of the leaf(2.03~5.00 mg·g^(-1)),and the branch(1.57~3.79 mg·g^(-1))was the lowest.Conclusion This method is rapid,sensitive,accurate and reliable,and can be used to determine the content of isovinoside lactone in Nauclea officinalis Pierrc ex Pitard,and provide a reference for its quality control.
