摘 要:乙醛脱氢酶2(ALDH2)是人类肝脏中氧化乙醛的关键酶,能将乙醛转化为乙酸,在酒精的代谢过程中,ALDH2发挥着重要的解毒作用。当ALDH2的酶活性受其编码基因ALDH2多态性的影响而致酶活性下降时会导致乙醛在体内蓄积,增加癌症风险。越来越多的研究表明ALDH2的基因多态性与肿瘤的发生、发展密切相关。就ALDH2的基因多态性在胃癌、食管癌、肝癌、肺癌、其他癌症的发生、发展及药物治疗进行总结。Aldehyde dehydrogenase-2(ALDH2) is the key enzyme that oxidizes acetaldehyde in liver, which can convert acetaldehyde to acetic acid. ALDH2 plays an important detoxification role in the metabolism of alcohol. When the enzyme activity of ALDH2 is weaken by the polymorphism of ALDH2, it will lead to the accumulation of acetaldehyde in the body and increase the risk of cancer. A growing number of studies have shown that gene polymorphism of ALDH2 is closely related to the development and treatment of tumors. This article summarizes the gene polymorphisms of ALDH2 in the occurrence, development, and drug treatment of gastric cancer, esophageal cancer, liver cancer, lung cancer, and other cancers.
