摘  要:目的:基于网络药理学探讨二陈汤治疗腺样体肥大(AH)的潜在作用机制。方法:运用中医数据库与分析平台(TCMSP)以二陈汤为关键词对成分和作用靶点进行检索。通过GeneCard和OMIM数据库收集AH疾病靶点。运用R语言筛选交集靶点,使用Cytoscape3.8.0软件对交集靶点进行网络药理的可视化分析。运用Metascape平台进行KEGG通路富集分析和GO功能富集分析。结果:共得到105个活性化合物,342节点,2076条边,主要活性化合物大多为黄酮类化合物,具有很好的抗病毒、抗炎、抗氧化及免疫调节等作用,PPI网络筛选得到23个核心靶点,排名前10的关键靶点为AKT1、MAPK1、JUN、IL6、MAPK8、STAT3、MYC、MAPK3、EGFR及HSP90AA1,KEGG富集通路主要涉及IL-17信号通路、PI3K-Akt信号通路、NF-κB信号通路、凋亡信号通路最为相关。结论:二陈汤通过下调炎症因子表达、减少促炎因子和趋化因子活性、调节细胞增殖、干扰病毒复制以及下调ROS缓解炎症的作用多途径干预模式减轻腺样体肥大,其信号通路与IL-17信号通路、PI3K-Akt信号通路、NF-κB信号通路、凋亡信号通路最为相关。Objective:To explore the potential mechanism of Erchen decoction in the treatment of adenoid hypertrophy(AH)based on network pharmacology.Methods:Using the Traditional Chinese Medicine Database and Analysis Platform(TCMSP)to search for the ingredients and action targets with Erchen Decoction as the key word.AH disease targets were collected through the GeneCard and OMIM databases.The R language was used to screen the intersection targets,and the Cytoscape3.8.0 software was used to perform a visual analysis of the network pharmacology of the intersection targets.KEGG pathway enrichment analysis and GO functional enrichment analysis were performed using the Metascape platform.Results:A total of 105 active compounds,342 nodes,and 2076 edges were obtained.The main active compounds were mostly flavonoids,which have good antiviral,anti-inflammatory,antioxidant and immunomodulatory effects.PPI network screening yielded 23 cores.Targets,the top 10 key targets are AKT1,MAPK1,JUN,IL6,MAPK8,STAT3,MYC,MAPK3,EGFR and HSP90AA1,KEGG enriched pathways mainly involve IL-17 signaling pathway,PI3K-Akt signaling pathway,NF-κB signaling pathway and apoptosis signaling pathway are most relevant.Conclusion:Erchen decoction alleviates adenoid hypertrophy by down-regulating the expression of inflammatory factors,reducing the activities of pro-inflammatory factors and chemokines,regulating cell proliferation,interfering with virus replication and down-regulating ROS to relieve inflammation signaling pathway,PI3K-Akt signaling pathway,NF-κB signaling pathway,and apoptosis signaling pathway are th e most relevant.
