肠复方对肠癌荷瘤小鼠免疫调节作用的研究

摘  要:目的:探讨扶正祛邪(肠复方全方)、扶正(肠复方健脾益气扶正组分)、祛邪(肠复方祛湿化瘀解毒组分)三种治则对肠癌荷瘤小鼠瘤体免疫调节作用的影响。方法:建立肠癌CT26荷瘤小鼠模型,随机分为模型组、扶正祛邪组、扶正组、祛邪组、香菇多糖组,共5组。分别给予相应药物干预4周,干预结束后计算各组小鼠体质量、抑瘤率,流式细胞仪检测瘤体细胞悬液中NK细胞表达,免疫组化法检测瘤体调节性T细胞、转化生长因子β(transforming growth factors-β,TGF-β)、白细胞介素-2(interleukin-2,IL-2)、IL-10、肿瘤坏死因子-α(tumor necrosis factor alpha,TNF-α)表达。结果:与模型组比较,三种不同治则组及香菇多糖组对于瘤体的增长有明显的抑制作用(P<0.01,P<0.05),调节性T细胞、IL-10、TGF-β表达显著下调(P<0.01,P<0.05),IL-2、TNF-α表达明显上调(P<0.01,P<0.05),三种不同治则组NK细胞表达显著上调(P<0.01)。结论:肠复方三种不同治则均可抑制肠癌小鼠肿瘤生长,其机制可能与调节机体免疫微环境相关。Objective:To explore the effect of Immunomodulatory effects of 3 therapeutic principles of Fuzheng Quxie(Chang Fu Fang),Fuzheng(constituent part of invigorating the spleen,nourishing Qi and strengthening the body in Chang Fu Fang)and Quxie(constituent part of removing Dampness,removing Stasis and detoxification in Chang Fu Fang)on colorectal cancer of model mouse.Methods:The CT26 tumor-bearing mouse model of intestinal cancer was established and randomly divided into five groups:model group,Fuzheng Quxie group,Fuzheng group,Quxie group and Lentinan group.The corresponding drug intervention was given for 4 weeks.After the intervention,the body weight and tumor inhibition rate of the mice in each group were calculated.Flow cytometry was used to detect the expression of NK cells in the tumor cell suspension.Expression of transforming growth factors-β(TGF-β),interleukin-2(IL-2),IL-10,tumor necrosis factor-α(TNF-α).Results:Compared with the model group,the three different therapeutic components and the lentinan group had obvious inhibitory effects on tumor growth(P<0.01,P<0.05),regulatory T cells,IL-10,TGF-βThe expression was significantly down-regulated(P<0.01,P<0.05),the expression of IL-2 and TNF-αwas significantly up-regulated(P<0.01,P<0.05),and the expression of NK cells of the three different treatment components was significantly up-regulated(P<0.01).Conclusion:The three different treatment principles of intestinal compound can inhibit the tumor growth in mice with intestinal cancer,and the mechanism may be related to the regulation of the immune microenvironment of the body.
