摘 要:丝瓜是典型的短日照植物,光周期是影响其产量和品质的关键因素。CONSTANS(CO)是调控植物光周期成花转变的关键基因。本研究以海南主栽品种‘泰利大肉丝瓜’为研究对象,利用生物信息学技术对CO基因家族进行鉴定分析,并研究CO基因在不同光周期处理下的表达模式。结果表明,丝瓜中含有6个CO基因,编码的多肽链长度为321~402个氨基酸,蛋白分子量为34.95~46.54 kD,理论等电点为4.49~7.65。进化分析表明CO基因家族分为三类,每个类别蛋白的保守基元数量相同,基因中具有相似的内含子/外显子结构。LcCO蛋白均含有保守的CCT和B-box保守结构域。丝瓜LcCO启动子区域含有大量与光响应相关元件。利用实时荧光定量PCR技术发现LcCO1和LcCO5基因的表达与光周期反应呈正相关,LcCO3基因的表达与光周期反应呈负相关。本研究初步筛选出丝瓜开花生物钟调控的潜在基因,对调控丝瓜开花节律具有重要意义。Luffa is a typical short-day plant,and its photoperiod is the key factor affecting its yield and quality.CONSTANS(CO)is a key gene that regulates the photoperiod floral transition in plants.In this study,the Hainan main luffa variety'Taili darou'was used as the research object.Bioinformatics technology was used to identify and analyze the CO gene family,and the expression pattern of CO gene under different photoperiods was studied.The results showed that six CO genes were identified,encoded the peptides with the length of amino acidds ranged from 321 to 402,and the molecular weight ranged from 34.95 to 46.54 kD,and the theoretical isoelectric point was ranged from 4.49 to 7.65.Evolutionary analysis showed that the CO gene family was divided into three types,and the same number of conserved motifs and similar intron/exon structures were observed in each category.All LcCO proteins contain conserved CCT and B-box domains.The promoter regions of luffa CO genes contain a large number of cis-elements related to light response.The expression of LcCO1 and LcCO5 genes were positively correlated with the photoperiodic response,and the expression of LcCO3 gene was negatively correlated with the photoperiodic response by real-time fluorescence quantitative PCR.In this study,potential genes in the regulation of the flowering clock of luffa were screened out,which is of great significance for the regulation of the flowering rhythm of luffa.
