摘  要:ERF(Ehylene response factor)转录因子作为植物重要的转录调节基因,影响着植物整个生命周期的生长发育和植物抗逆。本研究从84K(Populus abla×P.glandulosa)中克隆出PtRAP2L14基因,生物信息学分析PtRAP2L14编码蛋白的跨膜结构和亚细胞定位,发现PtRAP2L14蛋白质在10~37 aa处有一个从外到内的跨膜螺旋,其中心位点为27 aa处,亚细胞定位预测发现该蛋白定位在细胞质中。本研究成功构建PtRAP2L14基因植物表达载体,并成功完成在84K杨中的遗传转化,获得阳性转基因株系9个,为深入研究PtRAP2L14基因在杨树生长发育及逆境胁迫中发挥的作用提供材料和理论基础。As an important transcription regulator gene,ERF(ethylene response factor)transcription factor affects the growth and development of plant life cycle and plant stress tolerance.In this study,the full-length cDNA sequence of PtRAP2L14 gene was cloned from 84K(Populus abla×P.glandulosa).The transmembrane structure and subcellular localization of PtRAP2L14 protein were analyzed by bioinformatics.The results showed that PtRAP2L14 protein had a transmembrane helix from the outside to the inside at 10~37 aa,and the heart site was 27 aa.The subcellular localization prediction showed that PtRAP2L14 protein was mainly in the cytoplasm.In this study,plant expression vector of PtRAP2L14 gene was successfully constructed,and genetic transformation was successfully completed in 84 K poplar.Nine positive transgenic lines were obtained,which provided material and theoretical basis for further study on the role of PtRAP2L14 gene in poplar growth and development and stress.
