摘  要:目的研究合欢皮对荷瘤小鼠疲乏、睡眠障碍及抑郁的影响及机制。方法建立Lewis肺癌小鼠模型,小鼠随机分为对照组、模型组及给药组。行为学实验用于评价小鼠的疲乏、抑郁程度,脑电图分析睡眠状态。采用化学方法、Hochest染色分别测定小鼠脑组织中谷氨酸的水平及脑组织的损伤程度。结合网络药理分析及动物实验结果,利用N2a细胞建立谷氨酸损伤模型,通过MTT、流式细胞术、蛋白免疫印迹等验证合欢皮对神经的保护作用。结果合欢皮可显著改善荷瘤小鼠自主轮跑、转棒停留及强迫游泳不动时间,睡眠破碎及睡眠时相比例失调状况得到显著改善,降低脑组织中谷氨酸的水平及凋亡;细胞实验证明合欢皮可抑制谷氨酸引起神经细胞的氧化、凋亡。结论合欢皮可显著改善肿瘤相关神经症状,抑制谷氨酸引起的神经损伤,有效成分主要集中在30%乙醇洗脱部分。OBJECTIVE To explore the effect and mechanism of Albiziae Cortex on fatigue,sleep disturbance and depression in tumor-bearing mice.METHODS A mouse model of Lewis lung cancer was established,and the mice were randomly divided into control group,model group,and A.Cortex group.Behavioral experiments were used to evaluate the fatigue and depression of mice,and EEG to analyze sleep status.Chemical methods and Hochest staining were respectively used to determine the level of glutamate in the brain tissue of mice and the degree of brain damage.Combined with the results of network pharmacological analysis and animal experiments,N2a cells were used to establish a glutamate injury model,and the protective effect of A.Cortex on nerves was verified by MTT,flow cytometry,and western blotting.RESULTS A.Cortex significantly improved the time of autonomous rotation,staying on rotating rods,and forced swimming immobility in tumor-bearing mice,and significantly improved sleep fragmentation and sleep phase imbalance,reduced glutamate levels and apoptosis in brain tissue,and cell experiments have proved that A.Cortex can inhibit the oxidation and apoptosis of nerve cells caused by glutamate.CONCLUSION The above results indicate that A.Cortex can significantly improve cancer-related neurological symptoms and inhibit nerve damage caused by glutamate,and the active ingredients were mainly concentrated in the 30%ethanol elution part.
