摘 要:目的探讨黄秋葵粗多糖对脾虚便秘模型小鼠的作用及机制。方法将60只昆明小鼠随机分为正常组、模型组、麻仁丸组(0.8 g·kg^(-1))及黄秋葵粗多糖低、中、高剂量组(0.4、0.8、1.6 g·kg^(-1))。采用灌胃番泻叶水煎液(每日0.8 mL)结合限制饮食饮水、饥饱失常的方法复制脾虚便秘模型小鼠。模型复制成功后,连续灌胃给药(15 mL·kg^(-1))7 d,每日观察小鼠状态。进行小鼠排便功能实验;测定血清中D-木糖含量;计算小肠墨汁推进率;采用免疫组化法测定小鼠肠组织酪氨酸激酶受体(c-kit)及血管活性肠肽(VIP)表达水平。结果与模型组比较,黄秋葵粗多糖中、高剂量组小鼠的首便时间显著缩短(P<0.01),6 h内排便数量、粪便质量及含水量均明显升高(P<0.05,P<0.01);墨汁推进长度、墨汁推进率及血清D-木糖浓度均明显升高(P<0.05,P<0.01);c-kit阳性细胞像素面积比值和H-score评分明显升高(P<0.01),VIP阳性细胞像素面积比值和H-score评分明显降低(P<0.01)。结论黄秋葵粗多糖对脾虚便秘小鼠具有治疗作用,可能与上调肠道组织中的c-kit表达水平及下调VIP表达水平有关。ObjectivePharmacodynamics and mechanistic study of Abelmoschus esculentus (L.) Moench crude polysaccharide on constipated model mice with spleen deficiency syndrome.MethodsSixty Kunming mice were randomly divided into normal group,model group,hemp seed pill(0.8 g·kg^(-1))group and Abelmoschus esculentus (L.) Moench crude polysaccharide low-, medium-and high-dose groups(0.4、 0.8、 1.6 g · kg^(-1)). Mice with spleen deficiency and constipation were replicated by gavage of Folium Sennae decoction(0.8 mL per day)combined with restricted diet and water intake, and irregular amount of eating. After the model was successfully replicated, the mice were continuously administered by gavage(15 mL·kg^(-1))for 7 days,and the status of the mice was observed daily. Mice defecation function experiment. The content of D-xylose in serum was determined. Small intestine ink advancement rate was calculated. The expression levels of tyrosine kinase receptor(c-kit)and vasoactive intestinal peptide(VIP)in the intestinal tissues of mice were determined by immunohistochemistry.ResultsCompared with the model group, first stool time of the mice in the medium-and high-dose groups of Abelmoschus esculentus(L.)Moench crude polysaccharide was significantly shortened(P<0.01),and the frequency of stools,stool quality and water content within 6 hours was significantly increased(P<0.05, P<0.01);the ink advancement length, ink advancement rate and serum D-xylose concentration were significantly increased(P<0.05, P<0.01);the c-kit positive cell pixel area ratio and H-score score were significantly increased(P<0.01),and VIP positive cell pixel area ratio and H-score score were significantly decreased(P<0.01).Conclusion Abelmoschus esculentus(L.)Moench crude polysaccharide has therapeutic effect on constipated model mice with spleen deficiency syndrome,which may be related to the up-regulation of c-kit expression level and down-regulation of VIP expression level in intestinal tissues.
