摘 要:目的:观察蒙药肉蔻-5味丸对抑郁模型大鼠海马组织Bcl-2、Bax蛋白表达的影响。方法:将50只雄性SD大鼠按体质量随机分为正常组,模型组,西药组,肉蔻-5味丸高、低剂量组,每组10只。采用慢性轻度不可预见性应激结合孤养方法建立大鼠抑郁模型,西药组大鼠给予氟西汀3.6 mg·kg^(-1)·d^(-1),肉蔻-5味丸高、低剂量组大鼠分别给予肉蔻-5味丸1.6、0.4 g·kg^(-1)·d^(-1),模型组和正常组大鼠给予等量蒸馏水,周期为28 d。糖水消耗实验和旷场实验观察大鼠行为学改变;利用免疫组化染色和Western Blot检测大鼠海马组织CA3区Bcl-2、Bax蛋白表达水平。结果:与正常组比较,模型组大鼠第7、14、21、28天糖水消耗率显著降低(P<0.05),旷场实验水平和垂直运动得分显著降低(P<0.01),海马组织Bcl-2蛋白表达及Bcl-2/Bax比值显著降低(P<0.01),Bax蛋白表达显著升高(P<0.05);与模型组比较,氟西汀组和肉蔻-5味丸高剂量组大鼠第14、21、28天糖水消耗率显著升高(P<0.05,P<0.01),旷场实验水平和垂直运动得分显著升高(P<0.01,P<0.05),海马组织Bcl-2蛋白表达及Bcl-2/Bax比值显著升高(P<0.05),Bax蛋白表达显著降低(P<0.05)。结论:肉蔻-5味丸发挥抗抑郁作用机制是可能与调控抑郁模型大鼠海马组织Bcl-2、Bax凋亡蛋白表达水平有关。Objective: To observe the effect of Mongolian medicine Roukou-5 Wei Wan on the expression of Bcl-2 and Bax protein in hippocampus of chronic unpredictable mild stress (CUMS) model rats. Methods: Fifty male SD rats were randomly divided into five groups according to their weight: normal group, model group, western medicine group, Roukou-5 Wei Wan high and low dose group, with 10 rats in each group. The depression model was established by CUMS combined with solitary feeding,rats in western medicine group were given fluoxetine 3.6 mg · kg;· d;, Roukou-5 Wei Wan high and low dose group were given Roukou-5 Wei Wan 1.6, 0.4 g · kg;· d;respectively, rats in model group and normal group were given equal amount of distilled water, the cycle was 28 days. The changes of depressive behavior were observed by sugar consumption and open field experiment;the expression levels of Bcl-2 and Bax in CA3 area of hippocampus were detected by IHC and Western Bolt. Results: Compared with the normal group, the sugar consumption level of model group at 7, 14, 21, and 28 days were significantly lower (P<0.05),the horizontal and vertical movement scores were significantly lower (P<0.01), the Bcl-2 protein expression and the Bcl-2/Bax ratio in hippocampus were significantly decreased (P<0.01), the Bax protein expression was significantly increased (P<0.05).Compared with the model group, the sugar consumption level at 14, 21, and 28 days of western medicine group and Roukou-5 Wei Wan high dose group were significantly increased (P<0.05, P<0.01), horizontal and vertical movement scores were significantly increased (P<0.01, P<0.05), Bcl-2 protein expression and Bcl-2/Bax ratio in hippocampus were significantly increased (P<0.05),Bax protein expression were significantly decreased (P<0.05). Conclusion: The antidepressant mechanism of Roukou-5 wei wan may be related to the regulation of Bcl-2 and Bax apoptosis protein expression and Bcl-2/Bax ratio in hippocampus of depression model rats.
