摘 要:目的:考察并比较茅苍术生品及麸炒品挥发油体外抗结肠炎的作用,基于IKB抑制蛋白激酶(IKK)/核转录因子κB(NF-κB)信号通路探讨其作用机制。方法:以脂多糖诱导人结肠上皮细胞炎症损伤模型,MTT法检测细胞活力,ELISA法检测炎症相关因子白细胞介素(IL)-4、IL-8、IL-12和肿瘤坏死因子-α(TNF-α)的水平;RT-PCR法检测IKKα、NF-κB、IL-4和IL-8 mRNA表达;Western blot法检测IKKα、p-IKKα、p-IKKβ、NF-κB蛋白表达量。结果:与模型组比较,茅苍术生品和麸炒品挥发油均能显著提高细胞的存活率(P<0.05);提高抑炎因子IL-4的水平(P<0.05),降低促炎因子IL-8、IL-12和TNF-α水平(P<0.05);提高IL-4 mRNA的水平(P<0.05),降低NF-κB和IL-8 mRNA水平(P<0.05);降低p-IKKα、p-IKKβ和NF-κB蛋白表达量(P<0.05);麸炒品的作用优于生品。结论:茅苍术生品及麸炒品挥发油具有明显体外抗结肠炎的作用,麸炒品的作用优于生品,其机制部分是通过调控IKK/NF-κB信号通路而发挥作用。Objective: To investigate and compare the effects of essential oil from crude and bran-processed Atractylodes lancea(AL) on colitis in vitro and its mechanism based on IKK/NF-κB signaling pathway. Methods: The inflammatory injury model of human colonic epithelial cells(HCoEpiC) by LPS was established. The cell viability was detected by MTT assay.The levels of interleukin(IL)-4, IL-8, IL-12 and tumor necrosis factor-α(TNF-α) were determined by ELISA, and the mRNA expressions of inhibitor of nuclear factor kappa-B kinase(IKK)α, nuclear factor-κB(NF-κB), IL-4 and IL-8 were determined by quantitative RT-PCR. The expressions of IKKα, p-IKKα, p-IKKβ, NF-κB proteins were determined by Western blot. Results: Compared with the model group, the cell viability were increased(P<0.05), and the production of IL-4was significantly increaed(P<0.05). But the productions of IL-8, IL-12 and TNF-αwere significantly decreaed(P<0.05).Furthermore, the mRNA expressions of IL-4 was significantly stimulated(P<0.05), but the mRNA expressions of IL-8 and NF-κB and the expressions of p-IKKα, p-IKKβ, NF-κB proteins were notably inhibited by essential oil obtained from both crude and bran-processed AL(P<0.05). The essential oil from the processed AL was more effective compared with that from crude AL.Conclusion: The volatile oil from crude and bran-processed AL have satisfactory effects on colitis in vitro, and the effect of branprocessed AL is better than that of crude AL. The anti-inflammatory effect of AL could partly be through IKK/NF-κB signaling pathway.
