摘 要:目的:从TLR4/MyD88/NF-κB信号通路探讨活血化瘀通络法治疗输卵管炎性不孕模型大鼠的作用机制。方法:将43只SPF级雌性SD大鼠采用混合菌接种法建立输卵管炎性不孕大鼠模型,随机分为模型组、活血化瘀组、通络组及活血化瘀通络组。另取10只正常大鼠为空白组。空白组和模型组予10 mL·kg;·d;蒸馏水,活血化瘀组、通络组、活血化瘀通络组分别予14.4 g·kg;·d;血府逐瘀汤、通络药、血府逐瘀汤加减合通络药,1次/d,药物灌胃28 d后处死大鼠,腹主动脉采血后留取输卵管组织;ELISA法检测血浆中IL-1β、IL-6及TNF-α含量;RT-PCR和Western Blot法检测输卵管组织TLR4、MyD88、NF-κB mRNA及相应蛋白的表达。结果:与模型组比较,活血化瘀组、通络组、活血化瘀通络组可显著降低血浆中IL-1β、IL-6及TNF-α表达量(P<0.05),显著降低输卵管组织中TLR4、MyD88 mRNA及蛋白表达(P<0.05);活血化瘀通络组各指标改善显著优于活血化瘀组和通络组(P<0.05)。结论:活血化瘀通络法可通过调节TLR4/MyD88/NF-κB信号通路发挥抗炎功效,治疗输卵管炎,优于单用活血化瘀法或通络法。Objective: To explore the mechanism of activating blood circulation to remove blood stasis and dredging collaterals in the treatment oftubal inflammatory infertile rat model by TLR4/MyD88/NF-κB signal pathway. Methods: A total of 43 SPF female SD rats were model for tubal inflammatory infertility by mixed bacteria inoculation and randomly divided into model group, HXHY,TL and HXTL, and 10 normal rats were taken as blank group. The contorl and model groups were given10 mL·kg;·d;distilled water, and 14.4 g·kg;·d;corresponding drugs in the HXHY, TL, HXTL group. The rats were killed by intragastric administration once a day for 28 days, and the oviduct tissues were taken after abdominal aorta blood collection;IL-1β, IL-6 and TNF-α expression in plasma were detected by ELISA;The expressions of TLR4, MyD88, NF-κB mRNA and proteins in fallopian tubes were detected by RT-PCR and Western Blot. Results: Compared with the model group, the expressions of IL-1β, IL-6 and TNF-α in plasma were significantly decreased in the treatment group (P<0.05). Significant decreased TLR4,My D88 mRNA and protein expression in fallopian tube tissues (P<0.05). The improvement of each index in the group of HXTL was significantly better than that in the group of HXHY and TL group (P<0.05). Conclusion: The method of activating blood to remove blood stasis and dredging collaterals has anti-inflammatory effect by regulating TLR4/MyD88/NF-κB signal pathway and treat salpingitis, which is better than HXHY or TL method alone.
