摘 要:目的:观察不同剂量黄芩-黄连煎剂(以下简称芩连煎剂)对2型糖尿病模型大鼠肝脏TLR4/NF-κB炎症信号通路的影响。方法:SPF级雄性SD大鼠予高脂高糖饲料联合链脲佐菌素(STZ)制备2型糖尿病大鼠模型,随机分为模型组,二甲双胍组(1.80 g/kg),芩连高、中、低剂量(10.20、5.10、2.55 g/kg)组,另设空白对照组。芩连高、中、低剂量组分别灌胃给予浓度为1、0.5、0.25 g/mL的芩连煎剂,二甲双胍组灌胃给予浓度为0.5 g/mL的二甲双胍溶液,空白对照组、模型组灌胃给予等体积0.9%氯化钠溶液。测定治疗前后大鼠体质量及空腹血糖;ELISA法检测肝脏炎症因子白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)浓度;Western Blot法检测肝脏TLR4、NF-κBp65蛋白水平。结果:与模型组比较,各治疗组大鼠体质量显著升高(P<0.01,P<0.05),空腹血糖水平显著降低(P<0.01),二甲双胍组和芩连高剂量组大鼠肝脏IL-1β、IL-6、TNF-α含量显著降低(P<0.01),二甲双胍组和芩连高、中剂量组TLR4、NF-κBp65蛋白表达降低(P<0.01,P<0.05)。结论:芩连煎剂可能通过抑制2型糖尿病大鼠肝脏TLR4/NF-κB炎症信号通路来实现降糖作用。Objective: To explore the effects of different doses Huangqin and Huanglian Herbs Decoction(hereinafter referred to as Qin-Lian Herbs Decoction) on the inflammatory signaling pathway TLR4/NF-κB in the liver of type 2 diabetes mellitus(T2DM)rats. Methods: Male SPF-grade SD rats were fed with high-fat and high-sugar fodder combined with ip STZ to prepare T2DM rat models. The model rats were randomly divided into model group, metformin(1.80 g/kg) group, and high, medium, and low dose(10.20,5.10, 2.55 g/kg) of Qin-Lian Herbs Decoction groups. There would prepare a blank control group. The Qin-Lian Herbs Decoction high,medium, and low dose groups were given crude herbs at a concentration of 1, 0.5, 0.25 g/mL. The metformin group was given Metformin solution at a concentration of 0.5 g/mL. The blank control group and the model group were given an equal volume of 0.9% saline. The body weight and fasting blood glucose of the rats were measured before and after treatment. IL-1β, IL-6, and TNF-α concentrations of liver tissues were detected by enzyme-linked immunosorbent assay. The protein levels of liver TLR4 and NF-κBp65 were detected by Western Blot. Results: Comparing with the model group, the body mass of rats in each treatment group increased significantly(P<0.01,P<0.05), and the level of fasting blood decreased significantly(P<0.01). Liver IL-1β, IL-6, TNF-α in metformin group and QinLian Herbs Decoction high dose group decreased significantly(P<0.01). The content of TLR4 and NF-κBp65 protein expression in the metformin group, Qin-Lian Herbs Decoction high and medium-dose group decreased significantly(P<0.01, P<0.05). Conclusion: QinLian Herbs Decoction may inhibit liver TLR4/NF-κB inflammatory signaling pathway to achieve a hypoglycemic effect.
