摘  要:目的:观察静宁颗粒对注意缺陷多动障碍(ADHD)模型大鼠生长状态和行为学的影响。方法:将50只雄性4周龄自发性高血压大鼠(SHR)随机分为模型组、静高组、静中组、静低组及哌甲酯组,每组10只。同时将10只同龄Wistar大鼠设为正常组。哌甲酯组给予盐酸哌甲酯混悬液1.5 mg/kg灌胃,静高、中、低组分别给予静宁颗粒混悬液23.14、11.57、5.785 g/kg灌胃,模型组和正常组给予等量0.9%氯化钠溶液灌胃。各组均每日灌胃1次,共6周。干预第5周进行旷场实验、第6周进行Morris水迷宫实验。结果:干预6周后,与模型组比较,静高组、哌甲酯组体质量增长显著下降(P<0.05)。旷场实验结果显示:静高组、静中组和哌甲酯组在运动路程、运动速度、中心区时间显著低于模型组(P<0.05),外周区时间显著高于模型组(P<0.05);静中组和哌甲酯组在运动时间显著低于模型组(P<0.05)。水迷宫空间探索实验结果显示:静高组、静中组和哌甲酯组在穿越平台次数和目标象限停留时间显著高于模型组(P<0.05);静高组和哌甲酯组目标象限运动距离显著高于模型组(P<0.05)。结论:静宁颗粒和哌甲酯均能够有效调控ADHD模型大鼠的学习记忆和运动能力。静宁颗粒在调控注意力方面优势更为明显,且对ADHD模型大鼠体质量增长的影响较小,而哌甲酯对多动、冲动行为的控制更优。Objective: To study the effects of Jingning Granule on growth state and behavior of attention deficit hyperactivity disorder(ADHD) model rats. Methods: Fifty male 4-week-old SHR rats were randomly divided into model group,Jingning Granule high-, medium-, and low-dose group, methylphenidate group according to random numbers, with 10 rats in each group. At the same time, 10 Wistar rats of the same age were set as the normal control group. Methylphenidate group was given 1.5 mg/kg methylphenidate suspension, Jingning Granule was given different concentrations of particle suspension according to different groups, the specific concentration was 23.14 g/kg high dose, 11.57 g/kg medium dose, 5.785 g/kg low dose, and the model group and normal group were given the same amount of normal saline. Each group was given once a day for 6 weeks.Open field test was conducted at the 5th week and Morris water maze test was conducted at the 6th week. Results: After 6 weeks of intervention, compared with the model group, the weight gain of Jingning Granule high dose group and methylphenidate group decreased significantly(P<0.05). The results of open field test showed that the Jingning Granule high-, and medium-dose group and methylphenidate group had lower movement distance, movement speed and central zone time than the model group(P<0.05),and the time in the peripheral zone was higher than the model group(P<0.05);Jingning Granules in the middle-dose group and methylphenidate group had lower exercise time than the model group(P<0.05). The results of the water maze space exploration experiment show that the Jingning Granule high-, medium-dose group and methylphenidate group cross the platform and the target quadrant residence time are higher than the model group(P<0.05). The target quadrant movement distance of the Jingning Granule high-dose group and methylphenidate group was higher than that of the model group(P<0.05).
