黄芪注射液与生脉注射液对气阴两虚证候细胞模型代谢的影响

摘  要:目的:探索建立体外血清诱导2型糖尿病气阴两虚证候细胞模型,探索证候模型细胞代谢物的变化。方法:建立血清诱导气阴两虚证候人肝癌细胞(HepG2)细胞模型;观察药物对细胞活性的影响;给予黄芪注射液、生脉注射液和雪菊醇提物进行干预,以方测证辅助验证模型;采用1H-NMR代谢组学方法检测证候模型细胞中代谢物变化规律。结果:含0.5%气阴两虚2型糖尿病患者血清诱导24 h时可抑制细胞活性,给予益气养阴药物黄芪注射液与生脉注射液干预后促进证候模型细胞活性(P<0.05)。证候细胞中异亮氨酸、亮氨酸、脯氨酸等氨基酸、α-葡萄糖、β-葡萄糖及果糖等糖类、脂肪酸及甘油磷酯酰胆碱等代谢物含量发生变化(P<0.05)。结论:血清诱导证候细胞模型在一定程度上可以反映气阴两虚证,黄芪注射液与生脉注射液对气阴两虚证候细胞模型具有改善作用。Objective: To explore the establishment of an in vitro serum-induced cell model of type 2 diabetes with qi and yin deficiency syndrome, to explore the changes of cell metabolites in the syndrome model. Methods: Serum-induced qi-yin deficiency syndrome human hepatoma cell(HepG2) cell model was established;the effects of drugs on cell activity were observed;Huangqi Injection, Shengmai Injection and ethanol extract of Coreopsis tinctoria were given for intervention, and the syndrome was tested with prescriptions. Auxiliary verification model;1H-NMR metabolomics method was used to detect the changes of metabolites in syndrome model cells. Results: Serum containing 0.5% qi and yin deficiency type 2 diabetic patients could inhibit cell activity at 24 h after induction, and stimulated the cell activity of the syndrome model after the intervention of nourishing qi and nourishing yin drugs, Huangqi Injection and Shengmai Injection. Amino acids such as isoleucine, leucine and proline,carbohydrates such as α-glucose, β-glucose and fructose in syndrome cells, the contents of fatty acids and metabolites such as glycerophosphatidylcholine changed(P<0.05). Conclusion: Serum-induced syndrome cell model can reflect qi-yin deficiency syndrome to a certain extent. Huangqi Injection and Shengmai Injection can improve the qi-yin deficiency syndrome cell model.
