摘 要:目的:比较研究南方菟丝子和金灯藤的化学成分,对差异性成分进行定量。方法:采用基于UPLC-QTOFMS/MS的代谢组学方法对南方菟丝子和金灯藤中小分子化学轮廓进行表征,利用多变量数据统计方法发现各自的特征性成分,整合HPLC-UV方法对45批南方菟丝子和金灯藤商品药材差异性成分进行定量分析。结果:南方菟丝子和金灯藤化学轮廓有明显差异,南方菟丝子特征性成分为金丝桃苷、紫云英苷、异槲皮苷和山奈酚,金灯藤特征性成分为咖啡酸、3,4-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸和绿原酸;8个差异性成分定量表明,南方菟丝子中黄酮类成分为金灯藤的21.5倍,金灯藤中有机酸成分是南方菟丝子的2.8倍;34批南方菟丝子中金丝桃苷含量均达到2020版药典标准要求,但11批金灯藤中金丝桃苷含量均未达到药典标准要求。结论:法定品种南方菟丝子和混淆品金灯藤的小分子化学轮廓、特征性成分及其含量均有明显差异,金灯藤作为菟丝子基原使用的合理性仍需药效等多方面的系统研究。Objective:To qualitatively and quantitatively compare the chemical profiles of the two origins of Cuscutae Semen. Methods:An ultra-high performance liquid chromatography/quadrupole time-offlight mass spectrometry(UPLC–QTOF-MS/MS)based metabolomics approach was established to compare the chemical profiles of Cuscuta australis. R. B(CA)and C. japonica Choisy(CJ). Chemometric approach was applied for exploring the characteristic chemical markers in CA and CJ. Then,the chemical markers of the two origins were quantified in 34 batches of CA and 11 batches of CJ by HPLC-UV. Results:CA and CJ had different chemical profiles. Totally,22 components were identified in CA and CJ.Compared with CJ,4 flavonoids including kaempferol,isoquercitrin,astragalin and hyperoside,could be regarded as chemical markers of CA. Whereas 4 organic acids,consisting of caffeic acid,3,4-di-caffeoylquinic acid,4,5-di-caffeoylquinic acid,chlorogenic acid were be considered as chemical markers in CJ. Quantitative results showed that the contents of total flavonoids in CA was 21.5-fold more than that in CJ. Whereas the total organic acid in CJ was 2.8-fold more than that in CA.Conclusion:CA and CJ had totally different chemical profile and chemical markers. And the contents of chemical markers in CJ was different from CA. Whether CJ could be used as potentially alternative origin of Cuscutae Semen or not need further pharmacological evaluation.
