摘 要:采用不同的培养方式对5株虫草进行培养,培养结束后对不同培养方式培养出虫草进行粗多糖提取,并对含量进行比较,为获得更多粗多糖而选择合适的培养基提供参考。采用液体培养基和固体(大米)培养基对几株虫草进行培养,培养完成后用热水浸提法(料液比1:80,温度90℃,提取时间3 h,重复提3次)对其进行粗多糖的提取,测定粗多糖含量,计算得率并进行比较。在采用固体(大米)培养和液体培养虫草后,对其(包括子实体、菌丝球、发酵液和大米培养基)提取粗多糖并计算得率。确定了相同的物种在固体(大米)培养基和液体发酵培养下的子实体、菌丝球以及两种不同培养基的粗多糖含量和得率均为固体培养高于液体发酵。实验用两种不同培养方式培养虫草提取粗多糖,计算得率比较哪种培养方式更适合获得更多虫草粗多糖。此方法目的明确、操作简便,可为获得更多粗多糖选择合适培养方式提供参考。Several Cordyceps were cultured in different media.After the culture,the content of crude polysaccharide in different media was determined and compared,so as to provide a reference for obtaining more crude polysaccharide and selecting the appropriate media.Methods:Several Cordyceps were cultured in liquid medium and rice medium.After culturing,the crude polysaccharide was extracted by hot water extraction method(material liquid ratio 1:80,temperature 90℃,extraction time 3 hours,repeated extraction 3 times).The content of crude polysaccharide was determined,the yield was calculated and compared.Results:after the Cordyceps was cultured in different ways,the crude polysaccharide was extracted from the culture medium and the yield was calculated.The results showed that the content and yield of crude polysaccharide in solid culture medium and liquid culture medium were higher than that in liquid culture.in this experiment,two different culture methods were used to extract crude polysaccharide from Cordyceps sinensis,and the calculated yield was better than which culture method was more suitable to obtain more crude polysaccharide from Cordyceps sinensis.The purpose of this method is clear and easy to operate,which can provide reference for obtaining more crude polysaccharides and selecting suitable culture methods.
