摘 要:目的:探究脐带间充质干细胞(human umbilical cord mesenchymal stem cells,HuMSCs)来源的外泌体(exosome)对脓毒症急性肾损伤大鼠的保护作用。方法:选取雄性SPF级SD大鼠30只随机分为对照组(Control组)、模型组(Model组)、MSC来源exosome处理组(Exosome组),每组10只。对比三组实验大鼠生存率、肾功能水平、血液炎性介质水平及肾组织增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)增殖情况。结果:与Model组相比,Exosome组肾脏血肌酐、尿素氮、胱抑素C水平均有明显下降,差异有统计学意义(P<0.05);Exosome治疗6 d后,大鼠血清IL-6、TNF-α和IL-1β显著下降(P<0.05)。PCNA增殖检测显示Model组增殖上皮细胞增多,Exosome组增殖细胞增多更加明显。结论:脐带间充质干细胞来源的外泌体对脓毒症急性肾损伤大鼠有保护作用,可改善肾功能、降低炎性水平和提高肾组织细胞增殖水平。Objective To explore the protective effect of exosomes derived from human umbilical cord mesenchymal stem cells(HuMSCs)on acute kidney injury in rats with sepsis.Methods Thirty SPF male SD rats were randomly selected and divided into 3 groups,the control group,the model group,and the MSC-derived exosme treatment group(Exosome group)with 10 rats in each group.The survival rate,the level of renal function,the level of blood inflammatory mediators and proliferating cell nuclear antigen(PCNA)in renal tissue were compared among the three groups.Results Compared with the Model group,the renal blood creatinine and urea nitrogen levels of the Exosome group were significantly reduced,and the difference was statistically significant(P<0.05).After 6 days of exosome treatment,serum IL-6,TNF-αand IL-1βdecreased significantly(P<0.05).PCNA proliferation test showed that the number of proliferating epithelial cells in model group increased,and the number of proliferating cells in the Exosome group was more obvious.Conclusion The exosomes derived from umbilical cord mesenchymal stem cells have protective effect on acute kidney injury in rats with sepsis,and can improve renal function,reduce the level of inflammation and increase the level of renal tissue cell proliferation.
