摘  要:根腐病是近年上升为广西地区大棚厚皮甜瓜生产上的重要病害,严重影响甜瓜产业的健康发展。为明确该病害病原菌的分类地位并获得抗病种质材料,对病原菌进行分离纯化和致病性测定,结合形态学和分子生物学特征对病原菌进行分类鉴定,并通过苗期抗病性鉴定方法筛选抗病种质材料。结果表明,致病菌株的菌落和分生孢子形态特征与前人报道的腐皮镰孢菌(Fusarium solani)一致。基于rDNA-ITS序列构建系统发育树,致病菌株与腐皮镰孢菌聚于同一最小分支,据此将广西厚皮甜瓜根腐病的病原菌鉴定为腐皮镰孢菌。从27份厚皮甜瓜种质材料中,筛选到抗性水平为高抗的材料3份,分别为M87、M102和M125;抗性水平为抗的材料6份,分别为M23、M78、M84、M100、M112和M123。明确了广西地区的根腐病病原菌种类,筛选出高抗根腐病的厚皮甜瓜种质资源,为选育抗根腐病的厚皮甜瓜新品种奠定了基础。Melon root rot has become an important disease in Guangxi in recent years, which seriously affects the healthy development of melon production. In this study, the pathogen was isolated and purified, the pathogenicity was tested, and identification was performed using both morphological and molecular characteristics. Furthermore, melon germplasm materials were screened for resistance by seedling tests. The results showed that the morphological characteristics of colony and conidia of the pathogenic strains were consistent with those of Fusarium solani. The phylogenetic tree constructed based on r DNA-ITS sequence show that the pathogenic strains and F. solani clustered in the same smallest branch. Therefore, the pathogen causing melon root rot in Guangxi was F. solani. Among 27 melon germplasm tested, M87, M102 and M125 showed high resistance level to the pathogen, and M23, M78, M84, M100, M112 and M123 showed resistant level.The pathogen species of melon root rot in Guangxi area was determined, and the resistant melon germplasm were identified in this study. Our research laid the foundation for the breeding of melon varieties resistant to root rot.
