摘 要:目的观察芪参益气滴丸对心肌梗死大鼠心脏的保护及对线粒体分裂、融合与Sirt1/Mfn1信号通路的影响。方法通过永久结扎SD大鼠左冠状动脉前降支建立心肌梗死模型,并将其随机分为模型组、芪参益气滴丸低剂量组(0.6 g/kg)、芪参益气滴丸高剂量组(1.2 g/kg),并另设空白对照组。每天灌胃1次,持续4周。分别在术后24 h、治疗2周和4周时采用超声心动图评价心脏功能;TTC染色观察心脏组织梗死范围;HE染色用于监测各组心肌组织病理变化;采用Western blotting检测Sirt1、Mfn1和Fis1蛋白在心脏中的表达水平。结果术后24 h,模型组和芪参益气滴丸组的收缩末期左心室内径明显大于空白对照组,且收缩末期左室前壁明显变薄。治疗2周后,芪参益气滴丸组舒张末期内径明显小于心肌梗死组(P<0.05);治疗4周后,芪参益气滴丸组左心室内径较模型组明显改善;与模型组相比,芪参益气滴丸组梗死范围更小,组织病理改变更少,Sirt1、Mfn1蛋白表达增加(P<0.05),Fis1蛋白表达下降(P<0.01)。结论芪参益气滴丸至少部分通过激活Sirt1/Mfn1信号通路,促进线粒体融合,衰减心肌梗死诱导的线粒体碎片化,改善心肌梗死后的心脏损伤。Objective:To observe the protective effect of Qishen Yiqi dripping pills on the heart of myocardial infarction rats,and the effect on mitochondrial fission,fusion and Sirt1/Mfn1 signaling pathway.Methods:MI model was established by permanent ligation of anterior descending branch of left coronary artery in SD rats.They were randomly divided into the model group(MI),Qishen Yiqi dripping pills low-dose group(0.6 g/kg),Qishen Yiqi dripping pills high-dose group(1.2 g/kg),and a blank control group.The rats in treatment groups were administered with Qishen Yiqi dripping pills once daily for four weeks.V entricular remodeling was evaluated by echocardiography at 24 h after surgery,2 and 4 weeks after Qishen Yiqi treatment,respectively.The infarct area of the heart were observed by TTC staining;HE staining was used to monitor the pathological changes of myocardial tissue in each group.The protein expression levels of Sirt1 and Mfn1 in myocardial tissue were monitored by Western blotting.Results:At 24 h after surgery,the end systolic left ventricular internal meridian of the MI group and the treatment groups was significantly larger than that of the control group,and the anterior wall of the left ventricle at the end of contraction was significantly thinner.After 2 weeks of treatment with Qishen Yiqi dropping pills,the end diastolic internal meridian was significantly smaller than MI group(P<0.05).After 4 weeks of treatment,the left ventricular diameter in the treatment groups was significantly improved compared with that in the MI group.Compared with the control group,MI group had a wider range of myocardial infarction,while the treatment groups had smaller myocardial infarction area.Compared with the MI group,the treatment groups had fewer infarcts,smaller histopathological changes,and increased Sirt1 and Mfn1 protein expression(P<0.05),while Fis1 protein expression decreased(P<0.01).Conclusions:Qishen Yiqi dropping pills can at least partially improve cardiac injury after myocardial infarction by activating the Sirt1/
