摘  要:目的通过观察沉默信息调节因子2相关酶1(SIRT1)/过氧化物酶体增殖物激活受体γ共激活因子1α(PGC-1α)的表达,探讨丹酚酸B和黄连素的复合物对糖尿病小鼠肾损伤的影响。方法雄性C57BL/6J小鼠经高脂饮食诱导2型糖尿病,将糖尿病小鼠随机分为模型组、丹酚酸B+黄连素组、黄连素组、丹酚酸B组。黄连素、丹酚酸B及其复合物灌胃8周后,观察各组小鼠肾脏组织病理学改变;比较各组小鼠血清肌酐(Scr)、血清尿素(Urea)、空腹血糖水平(FBG)的变化,以及各组小鼠肾脏组织SIRT1、PGC-1α的蛋白表达。结果与正常组小鼠相比,模型组肾脏病理可见肾小球肥大,系膜区增宽增厚等。丹酚酸B、黄连素及其复合物均可减轻糖尿病引起的肾脏微结构的改变,其中丹酚酸B和黄连素的复合物效果最为显著。不同药物治疗后,丹酚酸B+黄连素组Scr、Urea水平显着降低(P<0.05);丹酚酸B+黄连素组、黄连素组及丹酚酸B组肾脏组织SIRT1/PGC-1α的蛋白表达均有所上升(P<0.05),其中丹酚酸B+黄连素组效果最为显著。结论丹酚酸B和黄连素的复合物可显著改善糖尿病小鼠的肾损伤,该作用与其调控SIRT1/PGC-1α的表达有关,进而发挥对糖尿病肾病的防治作用。Objective:By observing the chemical expression of silent information regulator 1(SIRT1)/peroxisome proliferator-activated receptorγcoactivator-1α(PGC-1α)to explore the effects of the compound of salvianolic acid B and berberine on renal injury in diabetic mice.Methods:Male C57BL/6J mice were induced to type 2diabetes by a high-fat diet.Diabetic mice were divided into model group(MX),salvianolic acid B+berberine group(DH),berberine group(H),salvianolic acid group(D).After 8 weeks of intragastric administration of berberine,salvianolic acid B and their compound,the histopathological changes in the kidneys of mice in each group were observed.The changes in serum creatinine(Scr),serum urea(Urea),and fasting blood glucose(FBG)levels of mice in each group,as well as the protein expression of SIRT1 and PGC-1αin the kidney tissues of mice in each group were compared.Results:Compared with the mice in the normal group(ZC),kidney pathology in the MX group showed glomerular hypertrophy,widening and thickening of the mesangial area.Salvianolic acid B,berberine and their compound could alleviate the renal microstructure changes caused by diabetes,among which the effect of the compound of salvianolic acid B and berberine was the most significant.After treatment with different drugs,the levels of Scr and Urea in the DH group were significantly decreased(P<0.05).The protein expression of SIRT1/PGC-1αwas increased in the kidney tissues of groups DH,H and D(P<0.05),and the effect in group DH was most significant.Conclusion:The compound of salvianolic acid B and berberine can significantly ameliorate renal injury in diabetic mice,which may be associated with its regulation of SIRT1/PGC-1αexpression,and then exerts protective and preventive effects against diabetic nephropathy.
