摘 要:目的观察金荞麦水剂干预急性呼吸窘迫综合征大鼠后肺组织水肿及炎症性介质释放的变化。方法50只Wistar雄性大鼠随机分为空白组、模型组及金荞麦高剂量组、中剂量组、低剂量组,每组10只。除空白组外,其余大鼠均气管内滴注3 mg/kg的LPS 100μL制备急性呼吸窘迫综合征模型,两肺均匀分布LPS 6 h,开始第1次给药。空白组、模型组给予灌胃生理盐水6 m L/kg,金荞麦高、中、低剂量组分别灌胃金荞麦水剂12、6、3 mL/kg,早晚各给药1次,连续3 d。干预结束后采集各组大鼠腹主动脉血,进行血气分析观察氧合指数、检测血常规观察白细胞计数(WBC)、中性粒细胞百分率(NEU%);采集肺组织比较肺组织湿干重比(W/D);HE染色观察肺组织病理学形态;ELISA法检测肺组织中炎性介质释放情况。结果与模型组比较,金荞麦各剂量组WBC下降、NEU%下降、氧合指数升高(P<0.05);W/D比值、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)表达均下降(P<0.05);金荞麦高剂量组各项观察指标均优于中、低剂量组(P<0.05)。结论金荞麦可能参与调节急性呼吸窘迫综合征大鼠炎症介质释放,减轻肺水肿。Objective:To study the changes of pulmonary edema and the release of inflammatory mediators in rats with acute respiratory distress syndrome treated by Fagopyrum dibotrys injection.Methods:50 male Wistar rats were randomly divided into the blank group,model group,and high-dose,medium-dose,and low-dose groups of Fagopyrum dibotrys injection,10 rats in each group.Except for the blank group,the other rats were intratracheally instilled with 100μL of 3 mg/kg LPS to prepare the acute respiratory distress syndrome model.The blank group and model group were given intragastric administration of normal saline 6 m L/kg,and the high,medium and low dose groups were given 12,6,and 3 mL/kg of Fagopyrum dibotrys injections by intragastric administration,once in the morning and evening,for 3 consecutive days.After the intervention,abdominal aortic blood was collected from the rats in each group,blood gas analysis was performed to observe oxygenation index,blood routine was detected to observe white blood cells(WBC)and neutrophil percentage changes(NEU%);lung tissue was collected to compare the wet and dry weight of lung tissue ratio(W/D);HE staining was used to observe the pathological morphology of lung tissue;ELISA method was used to detect the release of inflammatory mediators in lung tissue.Results:Compared with the model group,the white blood cell count,the percentage of neutrophils and the oxygenation index of each dose group of Fagopyrum dibotrys injection decreased,and the difference was statistically significant(P<0.05);wet to dry weight ratio(W/D)ratio,tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and interleukin-10(IL-10)expressions were decreased(P<0.05);the observation indexes in the high dose group were better than those in the middle and low dose groups(P<0.05).Conclusion:Fagopyrum dibotrys injection may be involved in regulating the release of inflammatory mediators and reducing pulmonary edema in rats with acute respiratory distress syndrome.
