摘 要:目的通过网络药理学方法研究旋覆代赭汤治疗化疗性恶心呕吐(CINV)作用机制。方法利用TCMSP、SymMap、TCMID和TCM Database@Taiwan数据库筛选药物有效成分,利用Swiss Target Prediction数据库检索旋覆代赭汤靶点;通过GengCards、DisGeNet、Drugbank及OMIM数据库检索CINV靶点,并将药物和疾病靶点进行映射,通过Cytoscape软件可视化并筛选出关键靶点,对潜在治疗靶点进行GO和KEGG富集分析。结果旋覆代赭汤主要有效成分有119个,包含309个靶点。CINV靶点有830个,其中108个是潜在治疗靶点,肿瘤坏死因子(TNF)、白细胞介素-1β(IL-1β)、环加氧酶2(PTGS2)和基质金属蛋白酶2(MMP2)等为关键靶点。通过GO和KEGG富集分析发现旋覆代赭汤治疗CINV是通过HIF-1、PI3K/Akt和TNF等信号通路发挥止吐作用。结论通过网络药理学发现旋覆代赭汤治疗CINV的作用机制复杂,涉及多靶点、多通路,为今后研究提供了新思路。Objective:To study the mechanism of Xuanfu Daizhe Decoction in the treatment of CINV by network pharmacology.Methods:Databases such as TCMSP,SymMap,TCMID and TCM Database@Taiwan were used to screen the active ingredients.Swiss Target Prediction database was used to retrieve the targets of Xuanfu Daizhe Decoction.GengCards,DisGeNet,Drugbank and OMIM databases were used to retrieve the targets of CINV.The drug targets and disease targets were mapped.Cytoscape software was used to visualize and screen out the key targets.GO and KEGG enrichment analysis was performed on potential therapeutic targets.Results:There were 119 main effective ingredients in Xuanfu Daizhe Decoction,which contained 309 targets.CINV had 830 targets,108 of which were potential therapeutic targets,with TNF,IL1β,PTGS2 and MMP2 as key targets.Through GO and KEGG enrichment analysis,it was found that Xuanfu Daizhe Decoction exerted antiemetic mechanism through HIF-1,PI3K/Akt and TNF and other signaling pathways in the treatment of CINV.Conclusion:Through network pharmacology,it is found that the mechanism of Xuanfu Daizhe Decoction in treating CINV is complex,involving multiple targets and pathways,which provides a new idea for future research.
