摘  要:“的布”(DIBOA)是苯并噁嗪类化合物的一种,具有抗虫、抗菌、抗感等重要生物活性。该研究从高抗蚜虫的玉米自交系Mo17中克隆bx 8基因,并借助于生物信息学分析手段,对bx 8基因及其编码的酶的生物化学特性进行分析,结果表明:从Mo17中克隆的目标DNA序列长度为1380 bp;具有完整的开放阅读框(ORF),编码由459个氨基酸组成的蛋白,分子量为49601.59。与玉米标准基因组目标序列相比,从Mo17中克隆的目标DNA序列,存在4个碱基的突变,造成目的基因编码的蛋白质氨基酸序列中发生3个氨基酸的变异;该蛋白与参考蛋白均无信号肽,亲疏水结构极为相似,所以该蛋白是无信号肽、无跨膜区的膜内蛋白,存在23个磷酸化位点,与普通小麦、节节麦和野生二黎小麦几乎相似。该研究中克隆的基因所编码的蛋白质为bx 8基因的表达和功能验证奠定了基础。DIBOA is a kind of benzoxazine compounds,which has important biological activities such as anti-insect,anti-bacterial,and anti-sense.In this study,the bx 8 gene was cloned from a maize inbred line Mo17 with high resistance to aphids,and the biochemical characteristics of the bx 8 gene and its encoded enzyme were analyzed by using bioinformatics analysis.The results showed that the target DNA sequence from Mo17 was 1380 bp,which has a complete open reading frame and encodes a protein composed of 459 amino acids with a molecular weight of 49601.59.Compared with the maize standard genome,the sequence of the target DNA cloned from Mo17 had four base mutations,resulting in three amino acid mutation occurred in the amino acid sequence of the protein encoded by the target gene;no signal peptides were found in this protein and reference protein,which the hydrophilic and hydrophobic structures were similar,thereby revealing that this protein was an intramembrane protein without signal peptide and transmembrane region;this protein had 23 phosphorylation sites,which are almost similar to common wheat,Jiejie wheat,and wild Erli wheat.The protein encoded by the cloned gene laid a foundation for verification of bx 8 gene expression and function.
