摘 要:为研究bFGF对氧化应激状态下小鼠卵母细胞糖代谢水平的影响,以6~8周的雌性昆明小鼠为试验材料,获取的卵母细胞分别在正常培养液(正常组),含有250μmol/L过氧化氢(过氧化氢组)、150 ng/L bFGF+250μmol/L过氧化氢(抗氧化组)的培养液中培养20 h,结果表明:正常组、过氧化氢组以及抗氧化组3组细胞中的ROS、MDA、8OHdG水平面之间均有显著差异(P<0.05),同理3组细胞中的GSH、线粒体膜电位以及能荷水平间均具有显著差异(P<0.05),通过细胞糖代谢途径中关键酶的活性结果显示,过氧化氢组指标hk,pk,6-pfk酶活性显著高于其他2组(P<0.05),对照组与抗氧化组差异不显著,pdk1酶活性则相反。试验结果表明,bFGF对氧化应激状态下的卵母细胞有保护作用并且能够调节糖代谢能力,从而提高卵母细胞质量。In order to study the effect of bFGF on glucose metabolism of mouse oocytes under oxidative stress,female Kunming mice for 6-8 weeks were used as experimental materials.The oocytes were incubated in normal culture medium(normal group),containing 250μmol/L hydrogen peroxide(hydrogen peroxide group),150 ng/L bFGF+250μmol/L hydrogen peroxide(oxidation group)for 20 hours.The results showed that the levels of ROS,MDA and 8OHdG in the normal,hydrogen peroxide and antioxidant groups were significantly different(P<0.05),and the levels of GSH,mitochondrial membrane potential and energy charge in the three groups were also significantly different(P<0.05).The results of the activities of key enzymes in the glucose metabolism pathway showed that the activities of hk,pk,6 pfk in hydrogen peroxide group were significantly higher than the other two groups(P<0.05),while the differences between the control and antioxidant groups were not significant,and it was the opposite for pdk1 enzyme activity.The results showed that bFGF has a protective effect on oocytes under oxidative stress and can regulate glucose metabolism,thus improving oocyte quality.
