摘　　要：研究旨在观察杜仲皮水提液对大鼠成骨细胞BMP2基因表达的影响,为补肾中药治疗骨质疏松等骨科疾病提供基础数据,采用酶消化法分离大鼠颅骨成骨细胞,取第2代成骨细胞,加入不同浓度的杜仲皮水提液(3×10^(-1)~3×10^(-5) mg·mL^(-1)),提取细胞总RNA,采用实时荧光定量PCR法检测成骨细胞BMP2基因表达的变化。结果显示:杜仲皮水提液浓度为3×10^(-1) mg·mL^(-1)、作用24 h时对大鼠成骨细胞BMP2基因表达有显著的促进作用。说明杜仲皮水提液能够促进大鼠成骨细胞BMP2基因的表达,尤其是较高浓度(3×10^(-1) mg·mL^(-1))作用24 h时促进作用更加明显。The purpose of study was to observe the effects of the water extract from Eucommia ulmoides on the expression of BMP2 gene in rat osteoblasts,and to provide basic data for the treatment of osteoporosis and other orthopedic diseases with kidney tonifying Chinese medicine.Rat calvarial osteoblasts were isolated by enzymatic digestion,and the second-generation osteoblasts were taken and added with different concentrations of water extract from Eucommia ulmoides(3×10^(-1)-3×10^(-5) mg·mL^(-1))to extract the cells.Total cell RNA was extracted,and the changes of BMP2 gene expression in osteoblasts were detected by real-time polymorase chain reaction(PCR).The results showed that the water extract from Eucommia ulmoides at a concentration of 3×10^(-1) mg·mL^(-1) or for 24 h had a significant promoting effect on the expression of BMP2 gene in rat osteoblasts.It indicated that the water extract from Eucommia ulmoides could promote the expression of BMP2 gene in rat osteoblasts,especially when it was treated at a higher concentration(3×10^(-1) mg·mL^(-1))for 24 h.
