摘　　要：目的探讨铁调节蛋白2(IRP2)高表达对PC12细胞的细胞活力及铁调节蛋白1(IRP1)表达水平的影响。方法培养具有分泌多巴胺性能的PC12细胞,分别转染Vector空载质粒(Vector组)和携带IRP2基因的质粒(IRP2组)。通过MTT法检测两组细胞的存活率,通过免疫印迹法检测两组细胞中IRP1蛋白的表达情况。结果与Vector组相比,IRP2组细胞存活率显著降低(t=3.97,P<0.01);与Vector组相比,IRP2组IRP1蛋白的表达水平显著升高(t=2.59,P<0.05)。结论IRP2高表达导致PC12细胞存活率降低,同时导致细胞中的IRP1表达升高,但具体生理机制有待进一步研究。Objective To investigate the effect of the high expression of iron regulatory protein 2(IRP2)on PC12 cell viability and the expression of iron regulatory protein 1(IRP1)in PC12 cells.Methods PC12 cells which could secrete dopamine were cultured and transfected with empty vector plasmid(Vector group)or the plasmid carrying the IRP2 gene(IRP2 group).MTT assay was used to measure cell viability,and western blotting was used to measure the protein expression of IRP1 in both groups.Results Compared with the vector group,the IRP2 group had a significant reduction in cell viability(t=3.97,P<0.01)and a significant increase in the protein expression of IRP1(t=2.59,P<0.05).Conclusion The high expression of IRP2 may lead to a reduction in PC12 cell viability and an increase in the expression of IRP1,but further studies are needed to clarify the specific physiological mechanism.
