土元壳和卵鞘中甲壳素结构特征与比较分析

摘  要:本研究采用酸碱法从土元壳和孵化后的卵鞘中分离甲壳素。通过傅里叶变换红外光谱、x射线衍射、热重分析和扫描电镜分析对提取的昆虫甲壳素的理化性质进行测定,并进行比较不同材料来源的甲壳素物理结构的异同。结果表明,来源于土元壳的甲壳素与来源于卵鞘的甲壳素物理特性不同。土元壳甲壳素的红外光谱图谱显示酰胺带Ⅰ在1659和1628 cm-1处分裂为两个峰,卵鞘的在1633 cm-1处为单峰;XRD显示土元壳甲壳素具有较强峰,而卵鞘的峰较弱;热重分析显示土元壳甲壳素降解温度高于卵鞘的,这些特征显示土元壳甲壳素具有α-甲壳素特征,卵鞘的则具有β-甲壳素特征。这些结果将有助于昆虫甲壳素的进一步开发与应用。In this study,crude extract and chitin were isolated from Eupolyphaga slough and ootheca after hatching by acid-base method.The physical properties of the extracted insect chitin were determined using Fourier transform infrared spectroscopy,X-ray diffraction,thermogravimetric analysis and scanning electron microscopy and compared the properties of them.The results showed that the physical properties of the chitin from Eupolyphaga sloughs were different from those from oocytes.The IR spectra of the extracts from sloughs showed that the amideⅠwas split into two peaks at 1659 and 1628 cm-1,and the extracts from ootheca had one peak at 1633 cm-1.The XRD results show that the extracts from sloughs have strong peaks,and thermogravimetric analysis showed that the degradation temperature of extracts was higher than these from ootheca.These characteristics indicated that the extracts were chitin that crude extract and chitin were isolated from Eupolyphaga slough hadα-chitin structure,while these from ootheca hadβ-chitin characteristics.These results will contribute to the further development and application of insect chitin.
