摘 要:目的 探究益气活血化痰方对动脉粥样硬化小鼠粥样斑块消退的作用及机制。方法 72只SPF级ApoE^(-/-)小鼠随机分为空白对照组、模型组、阿托伐他汀组及益气活血化痰方低、中、高剂量组,空白对照组给予普通饮食喂养,其余5组给予高脂饮食喂养,建立动脉粥样硬化模型。给药14周后,通过HE、油红O、Masson染色观察主动脉斑块情况,全自动生化分析仪检测血脂水平,Western blot法检测主动脉LXRα、SREBP1、ABCG1、ABCA1蛋白表达,RT-qPCR法检测主动脉LXRα、SREBP1、ABCG1、ABCA1 mRNA表达。结果 与模型组比较,益气活血化痰方各剂量组能不同程度地降低小鼠血清TC、TG、LDL-C水平(P<0.05),升高HDL-C、APOA1水平(P<0.05)。益气活血化痰方能减少小鼠主动脉粥样斑块面积,降低脂质蓄积,减少胶原纤维水平(P<0.05);上调小鼠主动脉LXRα、SREBP1、ABCA1/G1蛋白和mRNA的表达。结论 益气活血化痰方可能通过调控机体胆固醇逆转运,缓解机体动脉粥样硬化斑块的发展进程,起到抗动脉粥样硬化作用。AIM To investigate the effects and mechanisms of Yiqi Huoxue Huatan Recipe(YHHR) on the regression of atherosclerotic plaques in mice.METHODS Seventy-two ApoE^(-/-) mice of SPF grade were randomly divided into normal group,model group,atorvastatin group,and low-dose,medium-dose and high-dose groups of YHHR.The normal group was fed with ordinary diet,and the other five groups were fed with high fat diet to establish atherosclerosis models.After 14 weeks of administration,the mice were subjected to the determinations of aortic plaque by HE,oil red O and Masson staining;blood lipid level by automatic analyzer;protein expressions of LXRα,SREBP1,ABCG1 and ABCA1 by Western blot;and mRNA expressions of LXRα,SREBP1,ABCG1 and ABCA1 by RT-qPCR.RESULTS Compared with the model group,YHHR groups decreased serum levels of TC,TG and LDL-C(P<0.05);increased levels of HDL-C and APOA1(P<0.05);alleviated aortic atherosclerotic plaques;and up-regulated protein and mRNA expressions of aortic LXRα,SREBP1 and ABCA1/G1.CONCLUSION The anti-atherosclerosis effects of YHHR may contribute to its efficacy in the mechanism of reverse cholesterol transport for reduction of atherosclerotic plaque.
