摘 要:目的 基于整合药理学探究当归四逆汤抗寒凝心脉型心肌梗死的作用机制。方法 采用中医药整合药理学研究平台(TCMIP) V2.0,检索并获取当归四逆汤组方活性成分及靶标、心肌梗死靶标信息,构建中药-成分-靶点-通路多维分析,得到蛋白质相互作用网络(PPI)。同时,通过基因本体数据库(GO)功能分析、京都基因和基因组百科全书库(KEGG)的通路富集分析,预测当归四逆汤抗急性心肌梗死的分子机制。结果 共筛选出与心肌梗死相关的靶标545个,中药活性成分1 661种,可通过调控赖氨酸和孕激素介导的卵母细胞、幽门螺杆菌感染中的上皮细胞信号传导、谷胱甘肽代谢、多种氨基酸和脂类代谢、逆转录失调等方面,对GTP酶正调控、中枢神经系统发育、神经肽信号通路、ERK1和ERK2级联负调控、磷脂酰肌醇生物合成的过程等进行通路干预。结论 当归四逆汤治疗寒凝心脉型心肌梗死时,主要涉及多条代谢途径及细胞外多种酶和因子的共同调控作用。AIM To explore the mechanism of Danggui Sini Decoction against myocardial infarction(AMI) due to Congealing Cold in Heart Vessels based on integrated pharmacology.METHODS The integrative pharmacology-based research platform of traditional Chinese medicine(TCMIP) V2.0 was used to retrieve and obtain information on the active ingredients of Danggui Sini Decoction and their targets,and myocardial infarction targets as well.The platform was used to construct a multi-dimensional analysis of traditional Chinese medicines-components-targets-pathways,and obtain the protein-protein interaction network(PPI).Meanwhile,gene ontology database(GO) functional analysis and Kyoto encyclopaedia of genes and genomes(KEGG)pathway enrichment analysis were used to predict the molecular mechanism of Danggui Sini Decoction against AMI.RESULTS A total of 545 targets related to myocardial infarction and 1 661 active ingredients of Danggui Sini Decoction had been identified.Pathway intervention could be performed on positive regulation of GTPase,central nervous system development,neuropeptide signaling pathway,negative regulation of the ERK1 and ERK2 cascade,and the process of phosphatidylinositol biosynthesis by regulating lysine,progesterone-mediated oocytes,epithelial cell signaling in Helicobacter pylori infection,glutathione metabolism,multiple amino acid and lipid metabolism,and improvement of transcription disorders,etc.CONCLUSION In the treatment of AMI due to Congealing Cold in Heart Vessels,Danggui Sini Decoction mainly involves multiple metabolic pathways and the common regulation of a variety of extracellular enzymes and factors.
