摘 要:采用转录组测序(RNA-seq)技术比较澳洲坚果在7个不同发育阶段的转录组,获得了大量差异表达的Unigene序列。通过GO(Gene Ontology)分类和代谢途径富集性分析,将这些差异表达的Unigene归类于包含脂肪酸生物合成途径在内的406个代谢途径。将Unigene序列在KEGG(Kyoto Encyclopedia of Genes and Genomes)数据库中进行比对,获得17个关键酶的同源蛋白序列。本研究通过编码这些同源蛋白的基因在澳洲坚果果仁脂肪酸合成的表达模式进行分析,可为获得澳洲坚果果仁中脂肪酸合成机制的解析提供理论参考,并为进一步的理论研究和澳洲坚果的遗传育种提供潜在的基因资源。The transcriptomes of Macadamia nut kernel at seven different developmental stages were compared by transcriptome sequencing(RNA-Seq),and then obtained a lot of differentially expressed unigenes.Through Gene Ontology(GO)classification and pathway enrichment analysis,all of these differentially expressed unigenes were classified into 406 metabolism pathways including fatty acid biosynthesis which is involved in lipid synthesis.Seventeen homologous proteins of key enzymes were obtained when the sequence were aligned against Kyoto Encyclopedia of Genes and Genomes(KEGG)database.Through analysis of expression profiles of these genes during the seven-stage of macadamia nut lipid synthesis,this research not only sheds light on the elucidation of macadamia nut lipid synthesis but also provides candidate genes for further theoretic researches and genetic.
