摘 要:目的探讨清解扶正颗粒(QFG)对宫颈癌Caski细胞株裸鼠皮下移植瘤生长的抑制作用及其机制。方法在12只雄性BALB/c裸鼠右前肢腋窝处接种Caski细胞,构建宫颈癌皮下移植瘤模型,待裸鼠右侧腋窝下出现绿豆大小移植瘤,即为模型建立成功。瘤体长至70~100 mm3后,将12只BALB/c裸鼠随机分为对照组和QFG组,每组6只。QFG组按5 mL/(kg·d)灌胃200 mg/mL QFG溶液,对照组灌胃同体积生理盐水,2组均1周连续给药6 d,每天1次,共给药6周。从给药开始,隔天监测2组裸鼠肿瘤体积和体质量;末次给药24 h后处死裸鼠,剥离瘤组织,称取瘤重并计算抑瘤率;TUNEL法检测瘤体组织的细胞凋亡,Western blot检测2组Bax、Bcl-2、PI3K、AKT和p-AKT蛋白表达情况并计算Bax/Bcl-2、p-AKT/AKT。结果与对照组比较,QFG组裸鼠在第27~41天瘤体体积明显降低(P均<0.05),而裸鼠体质量无明显差别(P均>0.05);与对照组比较,QFG组瘤重明显降低,抑瘤率明显增高(P均<0.05);与对照组比较,QFG组细胞凋亡率明显增高(P<0.05);与对照组比较,QFG组Bax蛋白表达水平增加,Bcl-2蛋白表达水平降低,Bax/Bcl-2明显增高(P均<0.05);与对照组比较,QFG组PI3K、p-AKT蛋白表达水平明显降低(P均<0.05),AKT蛋白表达水平无明显差别(P>0.05),但p-AKT/AKT降低(P<0.05)。结论QFG对人宫颈癌Caski细胞移植瘤生长具有明显抑制作用,其作用机制可能通过抑制PI3K/AKT通路与诱导细胞凋亡有关。Objective:To investigate the inhibitory effect of Qingjie Fuzheng Granule(QFG)on the growth of subcutaneous transplantation tumor of cervical cancer Caski cells in nude mice and its mechanism.Methods:The model of cervical cancer transplanted subcutaneously was established by inoculating Caski cells into the axilla of the right forelimb of male BALB/c nude mice.The model was established successfully when the mung bean size transplanted tumor appeared in the right axilla of the nude mice.After the tumor grew to 70~100 mm;,BALB/c nude mice were randomly divided into control group(n=6)and QFG group(n=6).The QFG group was perfused with 5 mL/(kg·d)of 200 mg/mL QFG solution,and the control group was given the same volume of normal saline by gavage.Both groups were given continuous administration for 6 days a week,once a day for 6 weeks.From the beginning of administration,the tumor volume and body mass were monitored every other day.At 24 h after the last administration,the nude mice were killed.The tumor tissue was stripped and weighed to calculate the tumor inhibition rate;cellular apoptosis of the tumor tissue was detected by TUNEL staining;the protein expressions of Bax,Bcl-2,PI3K,AKT and p-AKT were detected by Western blot,and the ratio of Bax/Bcl-2 and p-AKT/AKT were calculated.Results:Compared with the control group,the tumor volume of nude mice in QFG group decreased significantly from 27 to 41 days(P<0.05),but the body weight of nude mice had no significant difference(P>0.05).Compared with the control group,the QFG group significantly decreased the tumor weight,the ratio of p-AKT/AKT,and the expressions of Bcl-2,PI3K and p-AKT protein(P<0.05),increased significantly the tumor inhibition rate,the apoptosis rate,the ratio of Bax/Bcl-2,and the expression of Bax protein(P<0.05),but the expression of AKT protein had no significant difference(P>0.05).Conclusion:QFG has a significant inhibitory effect on the growth of cervical cancer Caski cells transplantation tumor,and its mechanism may be related to the induction
