柳蘑的低极性化学成分研究

摘　　要：目的:对柳蘑的低极性成分进行提取、分离、鉴定,为柳蘑的药效物质及质量标准研究提供依据。方法:将柳蘑粉碎成粗粉,95%乙醇超声提取,合并提取液,回收溶剂得浓缩液,以水分散,乙酸乙酯萃取,萃取液回收溶剂得浸膏,浸膏以石油醚-丙酮梯度硅胶柱层析,收集石油醚-丙酮100∶1的洗脱物,以RP-HPLC法分离得单体,进行结构鉴定。结果:在四至六流分中分得4个单体,均为麦角甾类化合物,经核磁共振氢谱、碳谱等数据分析,并与文献进行对照鉴定,分别为(22E)-麦角甾-5,7,9(11),22-四烯-3β-醇(1)、麦角甾醇(2)、(22E,24R)-麦角甾-7,22-二烯-3β-醇(3)、(24S)-麦角甾-7-烯-3β-醇(4),4种化合物均为首次从该植物中分离得到。结论:柳蘑低极性成分主要为麦角甾类化合物,推测麦角甾类为柳蘑的药效成分之一。Objective: To extract, separate, and identify the chemical composition of the low polarity in pholiota adiposa, so as to provide basises for the study of effective substances as well as quality standards of pholiota adiposa. Methods: Pholiota adiposa was crushed into coarse powder, extracted with 95% ethanol, combined with the extract, recovered the solvent to obtain the concentrated solution, dispersed with water, extracted with ethyl acetate, and recovered the solvent to obtain the extract. The extract was separated by petroleum ether acetone gradient silica gel column chromatography. The eluent of petroleum ether acetone 100∶1 was collected, and the monomer was separated by RP-HPLC for structural identification. Results:Four monomers were separated from four to six streams, all of which were ergosterol compounds. After the data analysis of nuclear magnetic resonance hydrogen spectrum and carbon spectrum, and compared with the literature, the chemical structures were(22E)-ergosterol-5,7,9(11), 22-tetraene-3β-ol(1), ergosterol(2),(22E, 24R)-ergosterol-7, 22-diene-3β-ol(3),(24S)-ergosterol-7-ene-3β-ol(4). The four compounds were isolated from the plant for the first time. Conclusion:Low polar components of pholiota adiposa are mainly ergot steroid compounds. It is speculated that ergosterol is one of the medicinal composition.
